The synthesis of alpha-alkenyl alpha,beta-unsaturated ketones using germanium(II) salts is reported. Oxagermacycles derived from alpha,beta-unsaturated ketones with germanium(II) salts and aldehydes can be transformed into a-alkenyl a 6-unsaturated ketones. Ammonium salts promoted the elimination of Ge(II) species to afford the two classes of alpha-alkenyl alpha,beta-unsaturated ketones in good yields. The alpha-alkenyl alpha,beta-unsaturated ketones are precursors for multisubstituted heterocycles.

The feasibility of using activity-directed synthesis to drive antibacterial discovery was investigated. Copper nanoparticles supported on graphitic carbon nitride (Cu2O NPs@ g-C3N4) make a photoactive catalyst that has been developed for the preparation of ynone, aminoindolizines, and pyrrolo [1, 2-a] quinoline in mild and sustainable conditions. The catalyst is well characterized by various analytical techniques. The reaction can be completed using visible light more smoothly without the requirement of any other external sources of energy. The Cu2O NPs@ gC(3)N(4) material is found to be a more efficient, active, and reusable catalyst for the synthesis of ynone, aminoindolizines, and pyrrolo [1, 2-a] quinoline. Green chemistry metrics such as atom economy (AE) and E-factor are calculated for the reaction, and it is found that the present protocol is more facile and follows the green chemistry principles.

The synthesis of different metal pincer complexes coordinating to the chiral PNP ligand bis(2-((2R,5R)-2,5-dimethyl-phospholanoethyl))amine is described in detail. The characterized complexes with Mn, Fe, Re and Ru as metal centers showed good activities regarding the reduction of several prochiral ketones. Comparing these catalysts, the non-noble metal complexes produced best selectivities not only for aromatic substrates, but also for different kinds of aliphatic ones leading to enantioselectivities up to 99% ee. Theoretical investigations elucidated the mechanism and rationalized the selectivity.

Among the five complexes, complex 3 was new and the molecular structure of complex 3 was determined by single crystal X-ray diffraction. A proposed mechanism was discussed.

A generalized, simple and efficient transfer hydrogenation of unsaturated bonds has been developed using HBP in and various proton reagents as hydrogen sources. The substrates, including alkenes, alkynes, aromatic heterocycles, aldehydes, ketones, imines, azo, nitro, epoxy and nitrile compounds, are all applied to this catalytic system. Various groups, which cannot survive under the Pd/C/H-2 combination, are tolerated. Various phytochemicals have been reported to protect against oxidative stress. However, the mechanism underlying has not been systematically evaluated, which limited their application in disease treatment. Nuclear factor erythroid 2 – related factor 2 (Nrf2), a central transcription factor in oxidative stress response related to numerous diseases, is activated after dissociating from the cytoskeleton – anchored Kelch – like ECH – associated protein 1 (Keap1). The Keap1-Nrf2 protein-protein interaction has become an important drug target. This study was designed to clarify whether antioxidantive phytochemicals inhibit the Keap1-Nrf2 protein-protein interaction and activate the Nrf2-ARE signaling pathway efficiently. Molecular docking and 3D – QSAR were applied to evaluate the interaction effects between 178 antioxidant phytochemicals and the Nrf2 binding site in Keap1. The Nrf2 activation effect was tested on a H2O2 – induced oxidative – injured cell model. Results showed that the 178 phytochemicals could be divided into high – , medium – , and low – total -score groups depending on their binding affinity with Keap1, and the high total – score group consisted of 24 compounds with abundant oxygen or glycosides. Meanwhile, these compounds could bind with key amino acids in the structure of the Keap1 – Nrf2 interface. Compounds from high – total – score group show effective activation effects on Nrf2. In conclusion, phytochemicals showed high binding affinity with Keapi are promising new Nrf2 activators.

Chitosan copper (II) oxide nanocomposite was synthesized, characterized and used to synthesize [1,2,3] triazoles. Nanocomposite was characterized by using FTIR, XRD, FESEM, and EDS techniques, which reflected rough morphology. The powerful catalytic activity of hybrid nanocomposite was utilized to synthesize chalcones (3a-p) in relatively high yields (82%-98%) and multicomponent regio-selective cycloaddition of chalones, aryl halides (4), and sodium azide to afford the expected N-2-aryl[1,2,3]triazoles (5a-h) (80%-95% yield) rather than N-1-aryl(1,2,3]-triazoles (6a-h). The performance of nanomaterial was optimized by several variables. The capability of the nanocomposite was compared with previous work and the nanocomposite was found more efficient, economic and reproducible. The hybrid nanocomposite could be easily isolated form the reaction mixture and recycled four times without any significant loss of its catalytic activity. The reported catalyst is an inexpensive for good yields of the triazoles and may be used at industrial production for the reported compounds. (C) 2019 Published by Elsevier B.V.

Novel 2-(aryloxymethyl)aziridines and 2((3-aryl-1-phenylallyloxy)methyl)aziridine derivatives were prepared via ring-opening reaction of epoxides. The synthesized derivatives were characterized by using elemental analysis (EA), FT-IR, C-13 NMR, and H-1 NMR. The in vitro antitubercular activities of the synthesized compounds were evaluated against Mycobacterium tuberculosis H37Rv (MTB H37Rv) strain using MTT-MABA assay. All the aziridine derivatives exhibited improved persuasive antitubercular activity against MTB H37Rv in comparison with standard drugs. Among the tested compounds, 2-(naphthalene-1-yloxy) methyl aziridine (5b), 2-(naphthalene-2-yloxy)methylaziridine (5c), 2(m-tolyloxymethyl)aziridine (5e), 2-(3-(4-methoxyphenyl)-1-phenylalloxy)methylaziridine (12b) and 2-(3(2-chlorophenyl)-1-phenylallyloxy)methylaziridine (12c) revealed promising activity against MTh H37Rv. Specifically, compound 5b and 12 b showed three-times more active (MIC = 0.5 mu g/mL) than the standard drugs ethambutol (MIC = 1.56 mu g/mL) and ciprofloxacin (MIC = 1.56 mu g/mL). Phytoestrogens are dietary estrogens having similar structure as of estrogen. Some of these phytoestrogens are androgen receptor (AR) antagonists and exhibit preventive role in the prostate cancer. However, in androgen-independent prostate cancer (AIPC) the ARs were mutated (T877A, W741L, F876L, etc.) and these mutant ARs convert the antagonist to agonist. Our aim in this study is to find phytoestrogens that could function as an antagonist with wild and mutant ARs. The phytoestrogens were analyzed for binding affinity with wild and mutant ARs in agonist and antagonist conformations. The point mutations were carried out using Chimera. The antagonist AR conformation was modeled using Modeller. We hypothesize that the compounds having binding affinity with agonist AR conformation could not function as a full or pure antagonist. Most of the phytoestrogens have binding affinity with agonist AR conformation contradicting previous results. For example, genistein which is a widely studied isoflavone has known AR antagonist property. However, in our study, it had good binding affinity with agonist AR conformation. Hence, to confirm our hypothesis, we tested genistein in LNCaP (T877A mutant AR) cells by qPCR studies. The genistein functioned as an antagonist only in the presence of an androgen indicting a partial agonist type of activity. The in-vitro results supported our docking hypothesis. We applied this principle and found syringaresinol could function as an antagonist with wild and mutated ARs. Further, we carried out molecular dynamics for the hit molecule to confirm its antagonist binding mode with mutant AR. Communicated by Ramaswamy H. Sarma
