A facile hydrophosphonylation of alkenes by phosphites promoted by potassium carbonate was developed. The reaction features include easy handling, environmental friendliness, and avoidance of the use of strong bases. A variety of alkenes are tolerated in this reaction, with moderate to excellent yields.

A series of novel pyrazolo[3,4-d]pyrimidines was synthesised. Twelve synthesised compounds were evaluated for their anticancer activity against 60 human tumour cell lines by NCI (USA). Compound 7d proved prominent anticancer activity. It showed 1.6-fold more potent anti-proliferative activity against OVCAR-4 cell line with IC50 = 1.74 mu M. It also exhibited promising potent anticancer activity against ACHN cell line with IC50 value 5.53 mu M, representing 2.2-fold more potency than Erlotinib. Regarding NCI-H460 cell line, compound 7d (IC50 = 4.44 mu M) was 1.9-fold more potent than Erlotinib. It inhibited EGFR and ErbB2 kinases at sub-micromolar level (IC50 = 0.18 and 0.25 mu M, respectively). Dual inhibition of EGFR and ErbB2 caused induction of apoptosis which was confirmed by a significant increase in the level of active caspase-3 (11-fold). It showed accumulation of cells in pre-G1 phase and cell cycle arrest at G2/M phase.

Eight energetic salts with a gem-dinitro group based on 1,2,4-oxadiazole were developed. All of the structures were confirmed by infrared (IR) spectroscopy, H-1 and C-13 nuclear magnetic resonance (NMR) spectroscopy, and high-resolution mass spectrometry (HRMS), and single-crystal X-ray diffraction was used to determine the structures of the pivotal intermediate 3-(chlorodinitromethyl)-5-methyl-1,2,4-oxadiazole (3), the potassium salt (4) and the hydrazinium salt (7). All of them are insensitive to impact (>40 J) and have higher energy levels than trinitrotoluene (TNT).

Herein, the universal design of high-efficiency stimuli-responsive luminous materials endowed with mechanochromic luminescence (MCL) and thermally activated delayed fluorescence (TADF) functions is reported. The origin of the unique stimuli-triggered TADF switching for a series of carbazole-isophthalonitrile-based donor-acceptor (D-A) luminogens is demonstrated based on systematic photophysical and X-ray analysis, coupled with theoretical calculations. It was revealed that a tiny alteration of the intramolecular D-A twisting in the excited-state structures governed by the solid morphologies is responsible for this dynamic TADF switching behavior. This concept is applicable to the fabrication of bicolor emissive organic light-emitting diodes using a single TADF emitter.

The asymmetric Michael reaction of 3(2H) furanones and alpha, beta-unsaturated ketones was investigated, with the employment of a bulky chiral primary amine. It is reported that cheap and easily available Hg(OTf)(2) can efficiently mediate the Sakurai-Hosomi reaction of N-tert-butyloxycarbonyl (Boc) amino sulfones, aldehydes, and alpha-fluoroalkyl ketones using allyltrimethylsilane, with the catalyst loading down to 0.5-5.0 mol%, enabling the facile access to synthetically valuable homoallylic alcohols or amines, respectively. A chemoselective 1,4-allylation of alpha,beta-unsaturated enones is also achieved under the catalysis of 5.0 mol% Hg(OTf)(2). In most cases, Hg(OTf)(2) exhibited intriguing properties superior to those of commonly used metal Lewis acids for such reactions, suggesting that mercury catalysis is worthwhile to explore.

A group of novel 1,4-bis(3,5-dialkyl-4H-1,2,4-triazol-4-yl)bencene and 5-aryltriaz- 1-en-1-yl-1-phenyl-1H-pyrazole-4-carbonitrile derivatives have been successfully synthesized and characterized by spectroscopic analyses. The triazenes were obtained in moderate yield by a coupling reaction between 5-aminopyrazol-4-carbonitrile and aryl diazonium chlorides, and their structures were confirmed by detecting the CN functional group in both IR and C-13 NMR spectra. Conversely, the novel 1,4-bis(3,5-dialkyl-4H-1,2,4-triazol-4-yl)benzene derivatives were obtained using a previously unreported and facile pathway starting with p-phenylenediamine with selected triethyl orthoalkylates and then hydrazine monohydrate, followed by refluxing in triethyl orthoalkylates. The structure of the methyl derivative was confirmed by X-ray analysis. The synthesized compounds were tested and evaluated as antimicrobial agents.

Most topical classes of aryl, heteroaryl, and vinyl bromides were found to efficiently react with a variety of internal conjugated alkenes. Alzheimer’s disease is a degenerative brain condition that is the leading cause of dementia affecting millions of people around the world. Therapeutic development has focused on the problem of the loss of basal forebrain cholinergic function, as it is the only evidence responsible for brain neurodegeneration in patients with Alzheimer’s disease. Several attempts to improve cholinergic neurotransmission have been investigated by minimizing synaptic degradation of acetylcholine using acetylcholinesterase inhibitors. In the current study, we explore the designing of a new series of nicotinonitrile-coumarin hybrids as potential acetylcholinesterase inhibitors. The new hybrids were prepared utilizing pyridine-2(1H)-thiones as starting precursors. The in vitro acetylcholinesterase (AChE) inhibitory activities were examined for the new nicotinonitrile-coumarin hybrid molecules, when compared with donepezil as a standard drug with IC(50)of 14 nM. Coumarin derivative, linked to 6-(4-nitrophenyl)-4-phenylnicotinonitrile, showed more effective inhibitory activity than the reference donepezil with IC(50)of 13 nM. The free radical-scavenging capabilities against DPPH of the new hybrid derivatives were screened. Additionally, their in vitro cytotoxic activities have been tested against various eukaryotic cells. Furthermore, docking study showed excellent interaction between nicotinonitrile-coumarin hybrids and AChE. [GRAPHICS] .

A method for the synthesis of derivatives of 8-hydrazino 6-thioxopyrano[3,4-c]pyridine has been developed. Derivatives of 8-hydrazino-6-(methylsulfanyl)pyrano[3,4-c]pyridines and 5-(or 3)-(alkylsulfanyl)pyrano[3,4-c][1,2,4]triazolo[4,3-a]pyridines have been synthesized. The thione-thiol tautomerism of 5-thioxopyrano[3,4-c][1,2,4]triazolo[4,3-a]pyridine has been studied. In a basic medium, the equilibrium shifts toward the thiol form, which has allowed synthesis ofS-alkyl-substituted triazolo[4,3-a]pyridines.
