The B(C6F5)(3)-catalyzed chemoselective hydrosilylation of alpha,beta- and alpha,beta,gamma,delta-unsaturated ketones into the corresponding non-symmetric ketones in mild reaction conditions is developed. Nearly 55 substrates including those bearing reducible functional groups such as alkynyl, alkenyl, cyano, and aromatic heterocycles are chemoselectively hydrosilylated in good to excellent yields. Isotope-labeling studies revealed that hexafluoro-2-propanol also served as a hydrogen source in the process.

An efficient approach to prepare trifluoromethyl-alpha-carbolines and ester group-substituted alpha-carbolines via the tandem cyclization reaction of 2-(2-aminophenyl)acetonitriles and trifluoromethyl 1,3-diones or beta,gamma-unsaturated alpha-ketoesters was reported. The transformation proceeded smoothly in the presence of catalytic environmental-benign iron salts, which are used to prepare the desired products in moderate to good yields.

Nano-ferrite-supported Cu nanoparticles (Fe-dopamine-Cu NPs) catalyzed anti-Markovnikov-selective hydroboration of alkenes with B(2)pin(2) is reported under mild reaction conditions. This protocol can be applied to a broad range of substrates with high functional group compatibility. In addition, we demonstrated the use of Fe-dopamine-Cu NPs as a catalyst for the beta-borylation of alpha,beta-unsaturated ketones and ester, providing alkylboronate esters in up to 98% yield. Various phytochemicals have been reported to protect against oxidative stress. However, the mechanism underlying has not been systematically evaluated, which limited their application in disease treatment. Nuclear factor erythroid 2 – related factor 2 (Nrf2), a central transcription factor in oxidative stress response related to numerous diseases, is activated after dissociating from the cytoskeleton – anchored Kelch – like ECH – associated protein 1 (Keap1). The Keap1-Nrf2 protein-protein interaction has become an important drug target. This study was designed to clarify whether antioxidantive phytochemicals inhibit the Keap1-Nrf2 protein-protein interaction and activate the Nrf2-ARE signaling pathway efficiently. Molecular docking and 3D – QSAR were applied to evaluate the interaction effects between 178 antioxidant phytochemicals and the Nrf2 binding site in Keap1. The Nrf2 activation effect was tested on a H2O2 – induced oxidative – injured cell model. Results showed that the 178 phytochemicals could be divided into high – , medium – , and low – total -score groups depending on their binding affinity with Keap1, and the high total – score group consisted of 24 compounds with abundant oxygen or glycosides. Meanwhile, these compounds could bind with key amino acids in the structure of the Keap1 – Nrf2 interface. Compounds from high – total – score group show effective activation effects on Nrf2. In conclusion, phytochemicals showed high binding affinity with Keapi are promising new Nrf2 activators.

Introduction: The root of Millettia speciosa Champ. (Leguminosae) is one of the well-known traditional Chinese medicines abundant in phenolic compounds and plays important roles in the treatment of pain or numbness of the joints, blood deficiency sallow, chronic bronchitis and chronic hepatitis. Objective: An ultra-high performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS/MS) based chemical profiling approach was established for the separation and characterization of isoflavonoids in the roots of Millettia speciosa Champ. Methods: The roots of Millettia speciosa Champ. were prepared by ethanol extraction followed by further extraction with chloroform. The extracts were analyzed by UPLC-Q-TOF-MS in positive and negative electrospray ion modes at different Collision Energy (CE) values. Results: A total of 48 components were simultaneously detected, of which 38 components, including 21 isoflavones, 4 phenolic acids, 4 isoflavanones, 2 phenolic aldehydes, 2 flavanones, 1 aliphatic acid, 1 chalcone, 1 flavonol, 1 isoflavane and 1 pterocarpan were unambiguously identified or tentatively assigned based on the retention time, UV spectra, characteristic molecular ions, MS/MS fragmentation data and reference standards. The isomers of isoflavonoids were distinguished using accurate mass, the diagnostic fragmentations of C-ring and UV spectra. The major diagnostic fragment ions of isoflavonoids were observed and the corresponding fragmentation pathways were proposed. Conclusion: This investigation provides important analysis data for further quality control, pharmacological and toxicological research on Millettia speciosa Champ.

Strawberry is one of the most popular fruits because of its sensorial and functional properties. However, strawberry crop is sensitive to salt stress conditions. Despite plants have plasticity, high salt concentrations induce molecular, biochemical, and physiological responses in plants. There is evidence that the abscisic acid (ABA) hormone acts as a signaling molecule under stress conditions; however, the molecular mechanisms involved in the synthesis and homeostasis of ABA and in the induction of phytochemical antioxidant compounds under stress conditions remain unclear. In this study, the effect of stress induced by NaCl (salt stress – SS), with or without the simultaneous application of ABA, on the ABA, phenylpropanoids and L-ascorbic acid (AsA) metabolisms were evaluated. The physiological aspects (Na, Cl and proline concentration, photosynthetic variables) were also studied. The results showed that salt stress increases the Na and Cl content in the leaves, affects photosynthetic variables and triggers the production of proline, pelargonidin-3-O-glucoside, total phenolic compounds and AsA content, alongside the upregulation of several genes from the phenylpropanoid and flavonoid pathways. These effects were accompanied by the induction of compounds and transcripts related to ABA biosynthesis, conjugation and catabolism. Otherwise, the exogenous application of ABA in salt stressed plants promotes a shift in gene expression and metabolism to mitigate the stress. Therefore, salt stress affects the metabolism of ABA, phenylpropanoids and AsA in strawberry increasing phytochemical composition which is strongly associated with an ABA-dependent mechanism.

An inexpensive, mild, and highly efficient epoxidation protocol has been developed involving bubbling SO2F2 gas into a solution of olefin, 30% aqueous hydrogen peroxide, and 4 N aqueous potassium carbonate in 1,4-dioxane at room temperature for 1 h with the formation of the corresponding epoxides in good to excellent yields. Bis(NHC) ligand precursors, L1, based on trans-1,2-diaminocyclohexane were designed and synthesized. To introduce chirality at the hydroxyamide side arm on the NHC of L1, a chiral beta-amino alcohol, such as enantiopure leucinol, was used. Cu-catalyzed asymmetric conjugate addition reactions of cyclic and acyclic enones with Et2Zn were selected to evaluate the performance of L1 as a chiral ligand. For the reaction of cyclic enone, a combination of [bis(trimethylsilyl)acetylene]-(hexafluoroacetylacetonato)copper(I) (Cu(hfacac)(btmsa)) with a (+/-)-trans-1,2-cyclohexanediamine-based bis(NHC) ligand precursor, (rac; S,S)-L1, which was prepared from (S)-leucinol, was the most effective. Thus, treating 2-cyclohexen-1-one (3) with Et2Zn in the presence of catalytic amounts of Cu(hfacac)(btmsa) and (rac; S,S)-L1 afforded (R)-3-ethylcyclohexanone ((R)-4) with 97% ee. Similarly, use of (rac; R,R)-L1, which was prepared from (R)-leucinol, produced (S)-4 with 97% ee. Conversely, for the asymmetric 1,4-addition reaction of the acyclic enone, optically pure (-)-trans-1,2-cyclohexanediamine-based bis(NHC) ligand precursor, (R,R; S,S)-L1, worked efficiently. For example, 3-nonen-2-one (5) was reacted with Et 2 Zn using the CuOAc/(R,R; S,S)-L1 catalytic system to afford (R)-4-ethylnonan-2-one ((R)-6) with 90% ee. Furthermore, initially changing the counterion of the Cu precatalyst between an OAc and a ClO4 ligand on the metal reversed the facial selectivity of the approach of the substrates. Thus, the conjugate addition reaction of 5 with Et2Zn using the Cu(ClO4)(2)/(R,R; S,S)-L1 catalytic system, afforded (S)-6 with 75% ee.

One-step synthesis of beta-carboxyl ketones from alpha,beta-unsaturated ketones using CO2 as a feedstock by photoelectro-synergetic catalysis is reported herein. In this ambient system, silica nanowires (SiNWs) electrode not only acts as the working electrode but also harvests extra light energy to promote the reaction.
