The transfer hydrogenation of ketones catalyzed by transition metal complexes has attracted much attention. A series of ruthenium(11) complexes bearing 2,6-bis(5,6-dialkyl-1,2,4-triazin-3-yl)pyridine ligands (R-BTPs) were synthesized and characterized by NMR analysis and X-ray diffraction. These ruthenium (II) complexes were applied in the transfer hydrogenation of ketones. Their different catalytic activity were attributed to the alkyl arms on the 2,6-bis(5,6-dialkyl-1,2,4-triazin-3-yl)pyridine. As the length of the alkyl arms rising, the catalytic activities of the complex catalysts decreased. By means of 0.4 mol % catalyst RuCl2(PPh3)(3-methylbutyl-BTP) in refluxing 2-propanol, a variety of ketones were reduced to their corresponding alcohols with >95% conversion over a period of 3 h. Fluxional chemical species such as bullvalene have been a valuable source of inspiration and fundamental insight into the nature of chemical bonds. A supramolecular analogue of bullvalene, i.e., a fluxional host-guest system, in which the ensemble of a well-defined host and guest is engaged in continuous, degenerate constitutional rearrangements, is still elusive, however. Here, we report experimental and computational evidence for guest-induced dynamic covalent rearrangements in the ammonium complexes of self-assembled orthoester cryptands. This unique behavior is made possible by the ammonium guest playing a dual role: it is sufficiently acidic to initiate dynamic covalent exchange reactions at the orthoester bridgeheads, and as a hydrogen bond donor it acts as a supramolecular template, governing the outcome of a multitude of possible intra- and intermolecular rearrangement reactions. One particularly striking example of inherent dynamic behavior was observed in host-guest complex [Na-4(+)subset of o-Me-2-2.1.1], which spontaneously rearranged into the larger and thermodynamically more stable complex [NH4+subset of o-Me-2-2.2.1], even though this process led to the formation of poor host o-Me-2-1.1.1 as a consequence of the excess of one subcomponent (diethylene glycol; 1 in our nomenclature). These inherently adaptive host-guest networks represent a unique platform for exploring the interrelationship between kinetic and thermodynamic stability. For instance, as a result of optimal NH4+ binding, complex [NH4+subset of o-Me-2-2.2.1] was found to be thermodynamically stable (negligible intermolecular rearrangements over weeks), whereas computational studies indicate that the compound is far from kinetically stable (intramolecular rearrangements).

Introduction Pulse crops are nutritious and therefore widely grown. Pulse seed coats are typically discarded, despite their high content of polyphenols that are known for their antioxidant properties and health benefits. A better understanding of polyphenol diversity and biochemical pathways will ultimately provide insight into how polyphenols are linked to health benefits, which will help to better utilise these seed coats. Objectives To explore polyphenol profiles among seed coats of diverse genotypes of five pulse crops using a targeted liquid chromatography mass spectrometry (LC-MS) method. Methods Four genotypes of each of common bean, chickpea, pea, lentil and faba bean seed coats were selected for analysis. Following extraction, polyphenols were quantified using LC-MS. Results An LC-MS method was developed to quantify 98 polyphenols from 13 different classes in 30 min. The low-tannin seed coats had the lowest concentrations of all polyphenols. Chickpea and pea seed coats had the most similar polyphenolic profiles. The black common bean showed the most diverse seed coat polyphenol profile, including several anthocyanins not detected in any of the other seed coats. Conclusion The LC-MS method reported herein was used to show polyphenol diversity within several polyphenol classes among the pulse crop seed coats. Detected in all seed coats, flavonols and hydroxybenzoic acids appear well-conserved in the edible Fabaceae. The presence of anthocyanins, flavan-3-ols and proanthocyanins in the coloured seed coats suggests that unique divergent branches were introduced in the flavonoid biosynthetic pathway, possibly in response to environmental stressors.

The synthesis of phytol-derived gamma -butyrolactones as well as their evaluation for deterrent activity towards peach-potato aphid Myzus persicae and antiproliferative activity against four selected cancer cell lines are reported. Products were obtained in good yields (19-96%) and their structures were fully characterized by spectroscopic data (NMR, HRMS). Four synthesized delta -halo-gamma -lactones (4-7) are new and have not been previously described in the literature. In the choice test phytol (1) appeared deterrent to M. persicae, whereas modifications of its structure did not cause the avoidance of the treated leaves by the aphids. In contrast, aphids were attracted to the leaves treated with the new trans-delta -chloro-gamma -lactone (6). Electrical Penetration Graph (EPG) technique applied to explore the aphid probing and feeding activity revealed that neither phytol nor lactone 6 affected aphid probing and the consumption of phloem sap, which means that both phytol and the lactone 6 might have acted as postingestive modifiers of aphid behavior. The results of in vitro antitumor assays showed that obtained phytol derivatives exhibit cytotoxic activity against studied cancer cell lines (leukemia, lung and colon carcinoma and its doxorubicin resistant subline). Halolactones 4-6 were identified as the compounds, which arrest cell cycle of leukemia cells mainly in G2/M and S phases.

Alzheimer’s disease is a degenerative brain condition that is the leading cause of dementia affecting millions of people around the world. Therapeutic development has focused on the problem of the loss of basal forebrain cholinergic function, as it is the only evidence responsible for brain neurodegeneration in patients with Alzheimer’s disease. Several attempts to improve cholinergic neurotransmission have been investigated by minimizing synaptic degradation of acetylcholine using acetylcholinesterase inhibitors. In the current study, we explore the designing of a new series of nicotinonitrile-coumarin hybrids as potential acetylcholinesterase inhibitors. The new hybrids were prepared utilizing pyridine-2(1H)-thiones as starting precursors. The in vitro acetylcholinesterase (AChE) inhibitory activities were examined for the new nicotinonitrile-coumarin hybrid molecules, when compared with donepezil as a standard drug with IC(50)of 14 nM. Coumarin derivative, linked to 6-(4-nitrophenyl)-4-phenylnicotinonitrile, showed more effective inhibitory activity than the reference donepezil with IC(50)of 13 nM. The free radical-scavenging capabilities against DPPH of the new hybrid derivatives were screened. Additionally, their in vitro cytotoxic activities have been tested against various eukaryotic cells. Furthermore, docking study showed excellent interaction between nicotinonitrile-coumarin hybrids and AChE. [GRAPHICS] .

The effect of Argentine propolis extracts against enzymes related to metabolic syndrome and oxidative stress, as well as the botanical origin of raw propolis, were studied. Histological and chemical analyses of propolis samples revealed that the botanical origin isZuccagnia punctata, an Argentine medicinal plant. The melissopalynological analysis showed both pollen grains ofZ. punctataand the other plant species. This result indicates that the differences found in the botanical remains compared to the palynological studies may have been caused by the bees selecting resinous shrubs mainly ofZ. punctatafor the production of propolis and other plants with flowers for the production of honey. The richness of propolis was remarkable in two flavonoid precursors (2 ‘,4 ‘-dihydroxy-3 ‘-methoxychalcone, 2 ‘,4 ‘-dihydroxychalcone), the major chemical components ofZ. punctata. The hydroalcoholic extracts of Argentine propolis andZ. punctatawere able to inhibit enzymes associated with the metabolic syndrome, including alpha-glucosidase, alpha-amylase and lipase, with IC(50)values between 7 to 14, 37 to 48, and 13 to 28 mu g/mL, respectively. Biological activity was mainly attributed to chalcones. Oxygen and nitrogen reactive species scavenging activity was determined by the assays of superoxide radical (O-2(center dot-)), hydroxyl radical (HO center dot), hydrogen peroxide (H2O2), nitric oxide (NO center dot), and cation radical (ABTS(center dot+)). Results showed SC(50)values between 115 to 278, 12.50 to 46; 39 to 92; 50 to 104.50 and 23 to 33.75 mu g/mL, respectively. This study suggests for the first time that propolis from Argentina is highly effective in inhibiting enzymes related to the metabolic syndrome and in free-radical scavenging that would justify its use as a dietary supplement or as a functional ingredient in special food. Practical Application Propolis from Catamarca, Argentina, is traditionally used as medicine and food. Its botanical origin isZuccagnia punctata, an endemic plant species popularly used as a medicine in Argentina. Propolis has the ability to regulate the activity of enzymes involved in the carbohydrate and lipid metabolism, and consequently in metabolic syndrome. Besides, its antioxidant capacity makes it a natural product that can be used as a dietary supplement or as a functional ingredient in special foods. It is important to highlight that in the Argentine Food Code, propolis was incorporated in 2008 as a dietary supplement and the present results give major added value to this product.

To develop novel simultaneous inhibition of PLK1 and BRD4 bromodomain by a single molecule, three series of novel pteridinone derivatives were designed, synthesized and evaluated for their biological activity. Most compounds exhibited moderate to excellent cytotoxic activity against A549, HCT116, PC-3 and MCF-7 cell lines. The most promising compoundIII(4)showed high antiproliferative effects on four cell lines with IC(50)values of 1.27 mu M, 1.36 mu M, 3.85 mu M and 4.06 mu M, respectively. A highly selective and efficient oxidative carbocyclization/borylation of enallenols catalyzed by palladium immobilized on amino-functionalized siliceous mesocellular foam (Pd-AmP-MCF) was developed for diastereoselective cyclobutenol synthesis. The heterogeneous palladium catalyst can be recovered and recycled without any observed loss of activity or selectivity.
