Three iridium(III) complexes (Ir(4tfmpq)(2)mND, Ir(4tfmpq())2mmND, and Ir(4tfmpq)2mpND) with the 4-(4-(trifluoromethyl)phenyl)quinazoline (4tfmpq) main ligand and 1,5-naphthyridin-4-ol derivatives (mND: 8-methy1-1,5-naphthyridin-4-ol, mmND: 2,8-dimethy1-1,5-naphthyridin-4-ol, mpND: 8-methy1-2-pheny1-1,5-naphth-yridin-4-ol) as ancillary ligands were studied. The complexes (Ir(4tfmpq)(2)mND, Ir(4tfmpq)(2)mmND, and Ir(4tfmpq())2mpND) emit pure red emissions of 626-630 nm with high photoluminescence quantum yields of 85.2-93.4% in CH2Cl2 and better electron mobilities than that of tri(8-hydroxyquinoline)aluminum. By employing three pure red emitters, all the phosphorescent organic light-emitting diodes exhibited superior performances with a maximum external quantum efficiency of 31.48% and the efficiency roll-off is very mild, which are among the best results ever reported for pure red organic light-emitting diodes using Ir(III) complexes. In addition, CIE(x, y) coordinates of (0.670, 0.327) are also close to the standard red emission required by the National Television System Committee.

A highly selective and efficient oxidative carbocyclization/borylation of enallenols catalyzed by palladium immobilized on amino-functionalized siliceous mesocellular foam (Pd-AmP-MCF) was developed for diastereoselective cyclobutenol synthesis. The heterogeneous palladium catalyst can be recovered and recycled without any observed loss of activity or selectivity. The high diastereoselectivity of the reaction is proposed to originate from a directing effect of the enallenol hydroxyl group. The resistance of tumors against anticancer drugs is a major impediment for chemotherapy. Tumors often develop multidrug resistance as a result of the cellular efflux of chemotherapeutic agents by ABC transporters such as P-glycoprotein (ABCB1/P-gp), Multidrug Resistance Protein 1 (ABCC1/MRP1), or Breast Cancer Resistance Protein (ABCG2/BCRP). By screening a chemolibrary comprising 140 compounds, we identified a set of naturally occurring aurones inducing higher cytotoxicity against P-gp-overexpressing multidrug-resistant (MDR) cells versus sensitive (parental, non-P-gp-overexpressing) cells. Follow-up studies conducted with the P-gp inhibitor tariquidar indicated that the MDR-selective toxicity of azaaurones is not mediated by P-gp. Azaaurone analogs possessing pronounced effects were then designed and synthesized. The knowledge gained from structure-activity relationships will pave the way for the design of a new class of anticancer drugs selectively targeting multidrug-resistant cancer cells.

A catalytic annulation is presented that provides straightforward, modular synthetic access to 3-substituted indanones from benzoic acids and alpha,beta-unsaturated ketones. It is catalyzed by a bimetallic Ir/In system and proceeds via hydroarylation followed by Claisen condensation and optional retro-Claisen deacylation. Background and Purpose Previous studies have shown that Creosote bush-derived nordihydroguaiaretic acid (NDGA) exerts beneficial actions on the key components of metabolic syndrome including dyslipidaemia, insulin resistance and hypertension in several relevant rodent models. Here, we synthesized and screened a total of 6 anti-hyperlipidaemic analogues of NDGA and tested their efficacy against hepatic lipid metabolism in a high-fructose diet (HFrD) fed dyslipidaemic rat model. Experimental Approach HFrD fed Sprague-Dawley rats treated with NDGA or one of the six analogues were used. Serum samples were analysed for blood metabolites, whereas liver samples were quantified for changes in various mRNA levels by real-time RT-PCR. Key Results Oral gavage of HFrD-fed rats for 4 days with NDGA analogues 1 and 2 (100 mg center dot kg(-1)center dot day(-1)) suppressed the hepatic triglyceride content, whereas the NDGA analogues 2, 3 and 4, like NDGA, decreased the plasma triglyceride levels by 70-75%. qRT-PCR measurements demonstrated that among NDGA analogues 1, 2, 4 and 5, analogue 4 was the most effective at inhibiting the mRNA levels of some key enzymes and transcription factors involved in lipogenesis. All four analogues almost equally inhibited the key genes involved in triglyceride synthesis and fatty acid elongation. Unlike NDGA, none of the analogues affected the genes of hepatic fatty acid oxidation or transport. Conclusions and Implications Our data suggest that NDGA analogues 1, 2, 4 and 5, particularly analogue 4, exert their anti-hyperlipidaemic actions by negatively targeting genes of key enzymes and transcription factors involved in lipogenesis, triglyceride synthesis and fatty acid elongation. These analogues have therapeutic potential.

Triazolopyridinone derivatives are of high value in both medicinal and material chemistry. However, the chiral or hindered triazolopyridinone derivatives remain an underexplored area of chemical space because they are difficult to prepare via conventional methods. Here we report an electrochemical rearrangement for the efficient synthesis of otherwise inaccessible triazolopyridinones with diverse alkyl carboxylic acids as starting materials. This enables the efficient preparation of more than 60 functionalized triazolopyridinones under mild conditions in a sustainable manner. This method is evaluated for the late stage modification of bioactive natural products, amino acids and pharmaceuticals, and it is further applied to the decagram scale preparation of enantiopure triazolopyridinones. The control experiments support a mechanism involving an oxidative cyclization and 1,2-carbon migration. This facile and scalable rearrangement demonstrates the power of electrochemical synthesis to access otherwise-inaccessible triazolopyridinones and may find wide application in organic, material and medicinal chemistry. Chiral and hindered triazolopyridinone derivatives are an underexplored area of chemical space mainly due to their challenging synthesis via classical methods. Here, the authors report an electrochemical rearrangement for the synthesis of triazolopyridinones using diverse, available alkyl carboxylic acids as starting materials.

Nicotinic acid hydrazide was incorporated into new 4,5-dihydro-5-hydroxy-3,5-diphenylpyrazol-1-yl derivatives. Compounds 6a-h were synthesized, and their antihyperlipidemic activity was evaluated in high cholesterol diet-fed rat model. Compounds 6e, 6f were found to decrease the levels of serum total cholesterol by 14-19% compared to control group. Total triglycerides were also reduced by 24-28% and LDL cholesterol by 16%. As expected from parent niacin, compounds 6e and 6f caused an elevation of HDL cholesterol by 33-41%. Docking study supported the ability of designed compounds to block NPC1L1 active site in a manner similar to that observed with ezetimibe.

The present study was planned to apply the nanotechnology to therapeutically proven flavonoid-rich fraction for obtaining a formulation with enhanced antihyperglycemic activity. The leaves were extracted in aqueous methanol and fractionated to yield a flavonoid-rich fraction. This fraction was formulated into an aqueous nanosuspension using PVA through the nanoprecipitation method. The developed formulations were characterized for physical parameters and optimum nanosuspension was evaluated against type 2 diabetes in high-fat diet-fed, streptozotocin-induced diabetic animals. The pharmacokinetic parameter and histopathology were assessed to determine the kinetic profile and tissue architectures post oral administration of selected nano suspension. The selected nanosuspension was also studied for stability under accelerated conditions. Flavonoidrich fraction was obtained using Kupchan partitioning. The developed nanosuspension PG-EA-NS-B showed optimum physical parameters. In hyperglycemia induced by streptozotocin, in high-fat diet-fed rats, it effectively restores the normal level of blood glucose in the first hour. The effects were comparable to standard Glibenclamide. In biochemical observations, PG-EA-NS-B, as well as PG-EA, showed beneficial effects on various hepatic and renal parameters. The pharmacokinetic parameters showed that nanosuspension enhances the absorption and decreases the metabolism of flavonoid actives. The selected nanosuspension and free flavonoid fraction showed rectification of histological changes. The nanosuspension was most stable in cool conditions. PVA based nanosuspensions can be a viable strategy for oral delivery of natural flavonoids, as it significantly affected the absorption and metabolism of flavonoids upon oral administration.
