Efficient chemoselective transfer hydrogenation of the C=C bond of alpha,beta-unsaturated ketones has been developed, using the iridium complexes containing pyridine-imidazolidinyl ligands as catalysts and formic acid as a hydrogen source. In comparison with organic solvents or H2O as solvent, the mixed solvents of H2O and MeOH are critical for a high catalytic chemoselective transformation. This chemoselective transfer hydrogenation can be carried out in air, which is operationally simple, allowing a wide variety of alpha,beta-unsaturated substrates with different functional groups (electron-donating and electron-withdrawing substituents) leading to chemoselective transfer hydrogenation in excellent yields. The practical application of this protocol is demonstrated by a gram-scale transformation.

Two chiral polymers based on binaphthol(BINOL) were synthesized. The polymer can be used as a chiral inducer to coordinate with ZnEt2 to form a recoverable self-supporting catalyst, which could be used in the asymmetric catalytic reaction of alpha, beta-unsaturated ketones. The target products of high yield and good enantioselectivity(e.e. value up to 99%) can be obtained under mild conditions. The catalytic activity was not reduced significantly after 4 or 5 cycles of recovery of the recovered polymers.

Plant viral diseases, known as plant cancer, with high contagiosity can substantially reduce crop quality and yield. To identify potential anti-tobacco mosaic virus (TMV) agents with different mechanisms, a series of novel alpha-aminophosphonate derivatives containing a chalcone moiety were designed and synthesized. Bioassay results revealed that some target compounds exhibited improved curative activity against TMV in vivo, and the EC50 value of compound B-3 was 356.7 mg L-1. The activities of the defensive enzymes POD and CAT from tobacco leaves treated with B-3 and B-17 showed that these target compounds could improve the photosynthetic ability of the leaves and activate plant host resistance against TMV infection. The binding constant between B-3 and TMV Coat Protein (CP) (2.51 x 10(8) M-1), calculated by the fluorescence titration experiment and docking results, revealed that B-3 has a strong interaction with TMV CP. Further docking analysis revealed that B-3 was embedded between two layers of the TMV CP, which was consistent with the 2:1 binding mode of TMV CP and B-3 determined by the binding affinity experiment. The TEM morphological study of TMV treated with B-3 and B-17 indicated that this series of target compounds may trigger the disassembly of TMV by interacting directly with TMV CP. This study provides new insight for the discovery of antiviral compounds with two different mechanisms of action.

We describe an efficient synthesis of 2,4-substituted pyrido[4,3-d]pyrimidin-5(6H)-ones, which involves the acid-promoted cyclization of cyano enamine 15 to afford 2,4-bis(methylthio)pyrido[4,3-d]pyrimidin-5(6H)-one 17 as a key intermediate. Selective displacement of the 4-methylthio group by a wide range of anilines followed by oxidation of the 2-methylthio group and subsequent substitution by amines enabled the synthesis of a variety of 2,4-disubstituted pyrido[4,3-djpyrimidin-5(6H)-ones. A highly selective and efficient oxidative carbocyclization/borylation of enallenols catalyzed by palladium immobilized on amino-functionalized siliceous mesocellular foam (Pd-AmP-MCF) was developed for diastereoselective cyclobutenol synthesis. The heterogeneous palladium catalyst can be recovered and recycled without any observed loss of activity or selectivity. The high diastereoselectivity of the reaction is proposed to originate from a directing effect of the enallenol hydroxyl group. Optically pure cyclobutenol synthesis was achieved by the heterogeneous strategy by using chiral enallenol obtained from kinetic resolution.

Direct selective desaturation of carbonyl compounds to synthesize alpha,beta-unsaturated carbonyl compounds represents an environmentally benign alternative to classical stepwise procedures. In this study, we designed an ideal CeO2-supported Pd(II)-on-Au nanoparticle catalyst (Pd/Au/CeO2) and successfully achieved heterogeneously catalyzed selective desaturation of cyclohexanones to cyclohexenones using O-2 in air as the oxidant. Besides cyclohexenones, various bioactive enones can also be synthesized from the corresponding saturated ketones under open air conditions in the presence of Pd/Au/CeO2. Preliminary mechanistic studies revealed that alpha-C-H bond cleavage in the substrates is the turnoverlimiting step of this desaturation reaction.

Selenophenolato hydrido iron(II) complexes 1-3 cis-[(H)(SeAr)Fe(PMe3)(4)] (Ar = C6H5 (1), p-MeOC6H4 (2) and o-MeC6H4 (3)) could catalyze transfer hydrogenation of aldehydes and ketones. Among the three complexes, catalyst 1 exhibited the highest catalytic activity. The catalytic reactions took place under very mild conditions, using isopropanol as solvent and hydrogen source, (BuONa)-Bu-t as base under 60-80 degrees C. This catalytic system has good tolerance for many functional groups, such as halides, C=C double bonds, nitro groups and cyano groups at the phenyl ring of the substrates.

We previously identified a pyridomorphinan (6, SRI-22138) possessing a 4-chlorophenyl substituent at the 5′-position on the pyridine and a 3-phenylpropoxy at the 14-position of the morphinan as a mixed mu opioid receptor (MOR) agonist and delta/kappa opioid receptor (DOR/KOR) antagonist with potent antinociceptive activity and diminished tolerance and dependence in rodents. Structural variations at the 5′- and 14-positions of this molecule gave insights into the structure-activity relationships for binding and functional activity. Subtle structural changes exerted significant influence, particularly on the ability of the compounds to function as agonists at the MOR. In vivo evaluation identified compound 20 (SRI-39067) as a MOR agonist/DOR antagonist that produced systemically active potent antinociceptive activity in tail-flick assay in mice, with diminished tolerance, dependence/withdrawal, reward liability, and respiratory depression versus morphine. These results support the hypothesis that mixed MOR agonist/DOR antagonist ligands may emerge as novel opioid analgesics with reduced side effects.

The cyclic five-membered frustrated phosphane/borane Lewis pairs 11 a, b featuring the bulky octaethylhydrindacenyl- (Eind) substituent or its mono-bromo derivative ((Br)Eind) at phosphorus are monomeric at room temperature. The reactive frustrated Lewis pairs (FLPs) cleave dihydrogen. The cyclic FLP 11 b ((Br)Eind) undergoes 1,2-P/B addition to ethylene to give the zwitterionic heteronorbornane derivative 14 b. It reacts similarly with the carbon-carbon double bond of norbornene. With a variety of organic pi-reagents, the cyclic FLP 11 b often undergoes reaction sequences reminiscent of the Alder-Rickert reaction: the cycloaddition reaction is followed by rapid cycloreversion to form new five-membered heterocyclic FLP products with extrusion of ethene.
