To a solution of tert-butyl (3S)-3- [[4- [2- [(5 -iodo-2-methyl- 1 -naphthyl)oxy]-3 – pyridyl]pyrimidin-2-yl]amino]piperidine-1-carboxylate (110 mg, 0.17 mmol) in CH3CN (2 mL) was added 1,3-propanesultam (209.1 mg, 1.73 mmol), copper iodide (1.6 mg, 0.01 mmol), N,N?-dimethyl-1,2-ethanediamine (7.6 mg, 0.09 mmol) and potassium carbonate (47.8 mg, 0.34 mmol). The mixture was purged with N2 and stirred at 80 C for 88 h. After cooling down, the mixture was filtered, concentrated, dissolved in ethyl acetate (60 mL) and washed with H20 (50 mL x 2). The organic phase was dried over anhydrous sodium sulfate and concentrated and the residue was purified by Prep-TLC (50% ethyl acetate in petroleum ether,Rf= 0.1) to yield 55 mg (51%) of the title compound as a brown oil. LCMS (ESI) [M+H]=631.0.

The combined organic layer was washed with brine, dried over Na2SO4 and the solvent was removed under reduced pressure. The crude product was purified by column chromatography to obtain 2-{[3-(2-methyl-l,3-dioxolan-2- yl)tricyclo[3.3.1.03’7]non-l-yl] methyljisothiazolidine 1,1-dioxide as a viscous liquid (0.69 g) in 70% yield, m/z (M+l) 342; 1H NMR (CDCl3) 300 MHz delta 4.04-3.92 (m, 4H), 3.30 (t, J= 6.8 Hz, 2H), 3.10 (t, J= 7.4 Hz, 2H), 2.94 (d, J= 14.6 Hz, IH), 2.87 (d, J= 14.6 Hz, IH), 2.40-2.27 (m, 4H), 1.88-1.72 (m, 2H), 1.72-1.55 (m, 4H), 1.55-1.38 (m, 4H), 1.27 (s, 3H).

To a stirred solution of KHMDS in THF (1 M, 0.410 ml_, 0.410 mmol) and THF (1 mL) at 0 C was added a solution of 1 , 1-dioxo-isothiazolidine (0.044 g, 0.35 mmol) in THF (0.5ml_). The reaction mixture was stirred for 30 minutes at 0 C and then 3-[4-(bromomethyl)-3-chloro-phenyl]-5- (trifluoromethyl)-1 ,2,4-oxadiazole (0.100 g, 0.293 mmol), prepared according to procedures described in example 1 using 3-chloro-4-methyl-benzonitrile as starting material, was added. The reaction mixture was stirred at 0C for 2 h. The reaction mixture was then quenched with water and warmed to RT. The mixture was extracted with ethyl acetate and the combined organic layers washed with brine, dried over MgSC , filtered and concentrated under reduce pressure. LC/MS (Method A) retention time = 1.07 minutes, 398 (M+OH)”. The mass observed only in the negative ionization and corresponded to that of a hydrated species.

A 40 mL scintillation vial was charged with 1 ,1-dloxo-isothiazolidine (115 mg, 0.949 mmol) and OMF (3 mL). The reaction mixture was cooled to 00C and sodium hydride (95 mg, 2.373 mmol) was added. The reaction was left to stir at room temperature for 20 min foliowed by the addition of 3-bromo~5-(chloromethyl)pyridine hydrochloride (277 mg, 1.139 mmol). The reaction was stirred at room temperature for 3 h, then quenched with water and extracted with ethyi acetate. The organic layer was washed with water twice, dried over sodium sulfate and concentrated in vacuo. The crude was purified by silica gei flash chromatography using heptane-ethyl acetate (6(MO, v/v) to afford 3-bromo-5-(1,1- dioxo-isothiazolidin-2?ylmethyl)?pyridine as a colorless oil. Example 15 6-[(1,1-Dioxido-1,2-thiazolidin-2-yl)methyl]-1-(2-methoxyethyl)-5-methyl-3-(2-phenylethyl)thieno[2,3-d]pyrimidine-2,4(1H,3H)-dione To a solution of 49 mul (0.668 mmol) of propane sultam in 1.5 ml of DMF were added 27 mg (0.668 mmol) of sodium hydride (60% suspension in mineral oil) and then the mixture was stirred at RT for 25 min (“Solution 1”). After 20 min, one third of Solution 1 was added at 0 C. The reaction mixture was stirred at 0 C., with addition of a further third of Solution 1 after 20 min and after 40 min of reaction time. After the last addition, the cooling bath was removed and the reaction mixture was stirred at RT for about 18 h. All the volatile constituents were then removed on a rotary evaporator. The remaining residue was separated into its components by means of preparative HPLC (Method 8). After concentration of the product fractions and drying under high vacuum, 41 mg (64% of theory) of the title compound were obtained. 1H-NMR (400 MHz, CDCl3, delta/ppm): 7.41-7.16 (m, 5H, partially obscured by the CHCl3 signal), 4.30 (s, 2H), 4.25-4.17 (m, 2H), 4.11 (t, 2H), 3.70 (t, 2H), 3.35 (s, 3H), 3.26-3.15 (m, 4H), 2.99-2.88 (m, 2H), 2.50 (s, 3H), 2.44-2.28 (m, 2H). 100 mg (0.231 mmol) of (¡À)-1-(4-bromophenyl)-7,8-dimethoxy-N,4-dimethyl-4,5-dihydro-3H-2,3-benzodiazepine-3-carboxamide (Example 49A), 33.6 mg (0.278 mmol) of 1,2-thiazolidine 1,1-dioxide (CAS [5908-62-3]), 64 mg (0.46 mmol) of potassium carbonate and 13 mg (0.023 mmol) of allylchloropalladium dimer (CAS [12012-95-2]) are charged in 3 ml of 2-methyltetrahydrofuran and the suspension is degassed with argon for 10 min. Then 39 mg (0.093 mmol) of di-tert-butyl(2?,4?,6?-triisopropylbiphenyl-2-yl)phosphane (CAS [564483-19-8]) are added, degassing with argon is carried out again, and the mixture is heated at 80 C. for 16 h. The crude mixture is filtered, and then the solvent is removed and the residue obtained is purified by preparative HPLC. This gave 32 mg (29% of theory) of the desired product as a solid. In another reaction vessel, 50 mg (0.122 mmol) of the compound from Example 182A were dissolved in 1 ml of DMF, and 350 mul of Mixture 1 were added dropwise at 0 C. After 2.5 h at 0 C., a further 350 mul of Mixture 1 were added. After a further 30 min at 0 C., the reaction mixture was separated directly into its components by means of preparative HPLC (Method 8). Concentration of the product fractions and drying under high vacuum gave 42 mg (69% of theory) of the title compound. 1H-NMR (400 MHz, CDCl3, delta/ppm): 4.57 (d, 2H), 4.23-4.14 (m, 2H), 4.08 (q, 2H), 3.40 (t, 2H), 3.31-3.19 (m, 2H), 2.74-2.57 (m, 2H), 2.53-2.38 (m, 2H), 1.26 (t, 3H). Example 2; 0.15 g), isothiazolidine 1,1-dioxide (46 mg), tris (dibenzylideneacetone)- dipalladium (0) (6 mg), 4,5-bis (diphenylphosphino)-9, 9-dimethylxanthene (10 mg), cesium carbonate (130 mg) and 1,4-dioxane (2 mL). The Schlenk tube was sealed with a teflon screwcap and the reaction mixture was stirred at 100 C for 12 h. The reaction mixture was allowed to cool to room temperature, diluted with dichloromethane (10 mL), filtered and concentrated in vacuo. The crude product was purified by flash chromatography (dichloromethane to 10% MeOH in dichloromethane) to give 50 mg of the free base of the title compound. To a solution of this material in dichloromethane (0.3 mL) was added HCI (0.087 mL, 1 M in Et20), the solvent evaporated in vacuo and the material thus obtained triturated with Et20 to give 52 mg of the title compound as a white solid. NMR (‘H, DMSO): 8 10.57 (bs, 1H), 8.27 (bd, 1H), 8.19 (d, 1H), 7.94 (t, 1H), 7.82 (d, 1H), 7.55 (d, 1H), 7.32 (d, 2H), 7.18 (d, 2H), 4.03 (dd, 1H), 3.72 (m, 3H), 3.60/3. 20 (bm, 8H), 3.45 (s, 3H), 2.75 (s, 3H), 2.41 (m, 2H), 2.25 (m, 2H), 2.14 (m, 1H), 1.66/1. 10 (t/m, 2H).
