The most red-shifted hemithioindigo photoswitches have been identified through systematic introduction of aryl units to a parent pyrrole hemithioindigo photoswitch. Increasing the size of the 5′-aryl substituent is ineffective at producing further redshifted chromophores. A second generation of 3′,5′-diarylated photoswitches which possess increased tunability is reported. The development of an N-heterocyclic carbene (NHC) catalysed intercepted dehomologation of aldoses is reported. The unique selectivity of NHCs for aldehydes is exploited in the complex context of reducing sugars. Examples of strong substrate governance for either intercepted dehomologation or a subsequent redox-lactonisation were identified and mechanistically understood. More importantly, it was shown that catalyst design allowed the tuning of the selectivity of the reaction with structurally unbiased starting materials towards either of the two scenarios.

Phytoestrogens are dietary estrogens having similar structure as of estrogen. Some of these phytoestrogens are androgen receptor (AR) antagonists and exhibit preventive role in the prostate cancer. However, in androgen-independent prostate cancer (AIPC) the ARs were mutated (T877A, W741L, F876L, etc.) and these mutant ARs convert the antagonist to agonist. Our aim in this study is to find phytoestrogens that could function as an antagonist with wild and mutant ARs. The phytoestrogens were analyzed for binding affinity with wild and mutant ARs in agonist and antagonist conformations. The point mutations were carried out using Chimera. The antagonist AR conformation was modeled using Modeller. We hypothesize that the compounds having binding affinity with agonist AR conformation could not function as a full or pure antagonist. Most of the phytoestrogens have binding affinity with agonist AR conformation contradicting previous results. For example, genistein which is a widely studied isoflavone has known AR antagonist property. However, in our study, it had good binding affinity with agonist AR conformation. Hence, to confirm our hypothesis, we tested genistein in LNCaP (T877A mutant AR) cells by qPCR studies. The genistein functioned as an antagonist only in the presence of an androgen indicting a partial agonist type of activity. The in-vitro results supported our docking hypothesis. We applied this principle and found syringaresinol could function as an antagonist with wild and mutated ARs. Further, we carried out molecular dynamics for the hit molecule to confirm its antagonist binding mode with mutant AR. Communicated by Ramaswamy H. Sarma

Safety of Chalcone. I found the field of Chemistry very interesting. Saw the article A Chemometric Analysis of Compounds from Native New Zealand Medicinal Flora published in 2019.0, Reprint Addresses Reynisson, J (corresponding author), Univ Auckland, Sch Chem Sci, Private Bag 92019, Auckland 1142, New Zealand.. The CAS is 94-41-7. Through research, I have a further understanding and discovery of Chalcone.

Several hundred (396) compounds from New Zealand flora with medicinal properties were analyzed for their physicochemical properties. It was found that approximately 10% fulfilled all the requirements to be considered to be lead-like, over half of the compounds were deemed to be in the drug-like space and approximate to 75% were in the known drug space. These results indicate the presence of a significant proportion of compounds that are of particular interest to pursue as potential lead compounds or therapeutics. Additionally, compound classes were analyzed separatelymost carbonyl-containing compounds (aldehydes, ketones, esters and lactones), along with phenols were the most lead-like compounds, which also displayed very good proportions in the drug-like and known drug space. The information presented herein can be harnessed and utilized in future work, through focussing on the compounds and compound classes that exhibit high-levels of lead-likeness for further development.

The catalytic asymmetric epoxidation of alpha,beta-unsaturated ketones by tert-butylhydroperoxide (TBHP) has been well established using rare-earth metal amides RE[N(SiMe3)(2)](3) (RE = La(1), Nd(2), Sm(3), Y(4), Yb(5)) with chiral TADDOL ligands. It was found that the combination of Yb[N(SiMe3)(2)](3) and chiral TADDOL ligand H2L2 ((4S,5S)-2,2-dimethyl-alpha,alpha,alpha ‘,alpha ‘-tetra-3,5-bis(trifluormethylphenyl)-1,3-dioxolane-4,5-dimethanol) in a 1 : 1 molar ratio was the optimal choice, which provided the desired epoxides in excellent yields (89-99%) and good to high enantioselectivities (57-94% ee), using DBU as an additive. Various substrates were proved to have functional group tolerance. In addition, gram-level experiments and derivatization experiments were also studied.

A new methodology for the synthesis of alpha-diketones was reported via a one-pot cascade process from aldehydes and ketones catalyzed by a bifunctional iron nanocomposite using H2O2 as a green oxidant in water. An efficient and sustainable protocol for the hydration of alkynes has been developed under metal/acid/catalyst/ ligand-free conditions in a water/ethyl lactate mixture. The hydrogen-bond network in the ethyl lactate and water mixture plays a crucial and decisive role in activating the alkynes for hydration to afford the corresponding methyl ketones. This strategy gives the Markovnikov (ketone) addition product selectively over other possible products. The essential role of hydrogen bonding has been confirmed by experimental and theoretical techniques. A probable mechanism has been suggested by various control tests. The efficacy of the method has been further explored for the competent production of value-added alpha,beta-unsaturated carbonyl compounds through the reaction of aldehydes with alkynes as ketonic surrogates. The environmentally benign hydration method takes place under mild conditions, has broad functional-group compatibility, and uses the ethyl lactate/water (1:3) medium as a green alternative in the absence of any hazardous, harmful, or expensive substances. Rauhut-Currier (RC) reaction is an effective atom-economic method to construct C-C bond. However, the application of this reaction is limited by the lack of selectivity. Herein, an efficient intermolecular cross Rauhut-Currier reaction between chalcones and acrylates in the presence of the tributylphosphine catalyst was developed, the reactions were carried out in mild conditions, and performed well with a series of substrates, delivering the desired products with acceptable to good yields.
