Various phytochemicals have been reported to protect against oxidative stress. However, the mechanism underlying has not been systematically evaluated, which limited their application in disease treatment. Nuclear factor erythroid 2 – related factor 2 (Nrf2), a central transcription factor in oxidative stress response related to numerous diseases, is activated after dissociating from the cytoskeleton – anchored Kelch – like ECH – associated protein 1 (Keap1). The Keap1-Nrf2 protein-protein interaction has become an important drug target. This study was designed to clarify whether antioxidantive phytochemicals inhibit the Keap1-Nrf2 protein-protein interaction and activate the Nrf2-ARE signaling pathway efficiently. Molecular docking and 3D – QSAR were applied to evaluate the interaction effects between 178 antioxidant phytochemicals and the Nrf2 binding site in Keap1. The Nrf2 activation effect was tested on a H2O2 – induced oxidative – injured cell model. Results showed that the 178 phytochemicals could be divided into high – , medium – , and low – total -score groups depending on their binding affinity with Keap1, and the high total – score group consisted of 24 compounds with abundant oxygen or glycosides. Meanwhile, these compounds could bind with key amino acids in the structure of the Keap1 – Nrf2 interface. Compounds from high – total – score group show effective activation effects on Nrf2. In conclusion, phytochemicals showed high binding affinity with Keapi are promising new Nrf2 activators.

Axially chiral anilide compounds are an emerging but scarcely investigated class of stereogenic molecules with potential applications as biologically active scaffolds. Because of the lower rotation barriers, the synthesis of these compounds is a challenging task. Furthermore, the status of the limited structure type of chiral anilide constrains the latent capacity of the C-N axis as a chiral source in the application of asymmetric synthesis. Herein, we disclose an efficient protocol for the construction of the rationally designed axially chiral phosphamides via atroposelective N-allylic alkylation reaction of MBH carbonates and phosphamides. The simple hydroquinidine catalyst proves to be most efficient in this artroposelective strategy, delivering the desired axially chiral phosphamides in good yields and high enantioselectivities. In addition, a phosphamide compound, which contains both P-stereogenic center and C-N axial chirality, can be obtained by this method through a kinetic resolution process. Because of the large steric diaryl phosphoryl group, the synthesized axially chiral anilide has a large rotational barrier. As a demonstration, current studied axially chiral ortho-iodine substituted phosphamides could act as efficient chiral hypervalent iodine(III) catalysts for the asymmetric oxidative dearomatization of phenols. Moreover, a speculative model, which can explain the enantiocontrol, was proposed based on the experimental observation and theoretical calculation.

We report the highly enantioselective synthesis of P-chiral tertiary phosphine oxides featuring an ethynyl group via Cu(i)-catalyzed azide-alkyne cycloaddition. Newly developed chiral pyridinebisoxazolines (PYBOX) bearing a bulky C4 shielding group play an important role in achieving excellent enantioselectivity while suppressing side bis-triazoles formation in desymmetrizing prochiral diethynylphosphine oxides. Notably, by tuning the size of the C4 shielding group, it is possible to achieve excellent remote enantiofacial control in desymmetrizing phosphole oxide-diynes with the prochiral P-center farther from the ethynyl group by four covalent bonds. Time-dependent enantioselectivity is observed for these desymmetric CuAAC reactions, suggesting a synergic combination of a desymmetrization and a kinetic resolution, and our ligands prove to be better than unmodified PYBOX in both steps. This finding contributes to a highly enantioselective kinetic resolution of racemic ethynylphosphine oxides. The resulting chiral ethynylphosphine oxides are versatile P-chiral synthons, which can undergo a number of diversifying reactions to enrich structural diversity.

A one-pot, three-component [3 + 3] benzannulation reaction of alpha, beta-unsaturated carbonyl compounds, bromoallylic sulfones, and sodium sulfinates had been developed. A new heterogeneous acid catalyst based on a natural resource, Tunisian clay (Clay-H0.5), has been prepared and characterized by FT-IR, FE-SEM, and powder X-ray diffraction (XRD), as well as chemical composition, cation exchange capacity, specific surface area, and pore volume. Acid treatment for 0.5 h enlarged the surface area from 78.24 to 186.10 m(2)/g and pore volume (PV) from 0.186 to 0.281 cm(3)/g. The catalytic activity of this material was investigated in ketalization reaction under mild solvent-free conditions. This achieved up to 92% isolated yield for only 10 wt.% of the catalyst. This environmentally friendly method has advantages such as simple work-up procedure, avoidance of organic solvents, and good performance in ketalization reactions. Importantly, the Clay-H0.5 catalyst showed good recyclability where insignificant activity loss was exhibited even after six runs. Synthesized cyclic ketals were tested for their possible antileishmanial and antibacterial activities as well as antifungal activity. Biological screening showed that compound 11 had important antileishmanial activity against both L. major and L. infantum, while compound 14 also had significant antibacterial activity against four gram-positive and two gram-negative bacteria, and antifungal activity against Candida albicans, with minimal inhibitory concentration values ranging from 15.62 mu g/mL to 125 mu g/mL.

A chemoselective cascade cycloaddition reaction is developed for green and efficient access to cyclopenta-[c]pyridine derivatives. Simple and inexpensive NaOH is used as the sole catalyst for this process. The delta-carbon of cyclopropyl ester is activated as a nucleophiic carbon to initiate highly chemoselective cascade reactions. Cyclopenta-[c]pyridines bearing various substituents are afforded in excellent yields. Preliminary studies on the bioactivities of the afforded products show promising antibacterial activities for potential applications in plant protections.
