Aim: There is a continuous and urgent need for new anticancer agents with novel structures and target selectivity. Methods & results: The anticancer activity of the prepared compounds was assessed against human lung (A549) and stomach (AGS) cancer cell lines and evaluated in the noncancer human lung fibroblast (MRC-5) cell line. 2-Pyrazolines were devoid of toxicity in all cell lines used, chalcones bearing a beta-(benz)imidazole moiety being toxic toward AGS cell line. Mechanistic studies showed that these compounds trigger loss of cell viability and mitochondrial membrane potential, while eliciting morphological traits compatible with regulated cell death, which was ultimately shown to derive from caspase activation, specifically caspase-3. Conclusion: Chalcones 1-3 have been identified as new and promising anticancer agents toward the AGS cell line.

An oxidative esterification of aldehydes with alkanols catalyzed by an in situ generated low-valent cobalt system has been developed using an enone as a mild oxidant. Mechanistic studies revealed that it proceeds through a Co(i)-catalyzed hydrogen-transfer route, wherein the -vinyl moiety in the bidentate enone functions as a hydride acceptor. Meanwhile, Co(i)-catalyzed formyl C-H activation occurred as a competing reaction leading to aldehyde dimerization. The occurrence of the usually kinetically disfavored hydride transfer step therein was significantly increased in the presence of an enone reacting as a hydride transfer initiator.

The synthesis of alpha-alkenyl alpha,beta-unsaturated ketones using germanium(II) salts is reported. Oxagermacycles derived from alpha,beta-unsaturated ketones with germanium(II) salts and aldehydes can be transformed into a-alkenyl a 6-unsaturated ketones. Ammonium salts promoted the elimination of Ge(II) species to afford the two classes of alpha-alkenyl alpha,beta-unsaturated ketones in good yields. The alpha-alkenyl alpha,beta-unsaturated ketones are precursors for multisubstituted heterocycles.

The present study was planned to apply the nanotechnology to therapeutically proven flavonoid-rich fraction for obtaining a formulation with enhanced antihyperglycemic activity. The leaves were extracted in aqueous methanol and fractionated to yield a flavonoid-rich fraction. This fraction was formulated into an aqueous nanosuspension using PVA through the nanoprecipitation method. The developed formulations were characterized for physical parameters and optimum nanosuspension was evaluated against type 2 diabetes in high-fat diet-fed, streptozotocin-induced diabetic animals. The pharmacokinetic parameter and histopathology were assessed to determine the kinetic profile and tissue architectures post oral administration of selected nano suspension. The selected nanosuspension was also studied for stability under accelerated conditions. Flavonoidrich fraction was obtained using Kupchan partitioning. The developed nanosuspension PG-EA-NS-B showed optimum physical parameters. In hyperglycemia induced by streptozotocin, in high-fat diet-fed rats, it effectively restores the normal level of blood glucose in the first hour. The effects were comparable to standard Glibenclamide. In biochemical observations, PG-EA-NS-B, as well as PG-EA, showed beneficial effects on various hepatic and renal parameters. The pharmacokinetic parameters showed that nanosuspension enhances the absorption and decreases the metabolism of flavonoid actives. The selected nanosuspension and free flavonoid fraction showed rectification of histological changes. The nanosuspension was most stable in cool conditions. PVA based nanosuspensions can be a viable strategy for oral delivery of natural flavonoids, as it significantly affected the absorption and metabolism of flavonoids upon oral administration.

Nano-ferrite-supported Cu nanoparticles (Fe-dopamine-Cu NPs) catalyzed anti-Markovnikov-selective hydroboration of alkenes with B(2)pin(2) is reported under mild reaction conditions. This protocol can be applied to a broad range of substrates with high functional group compatibility. In addition, we demonstrated the use of Fe-dopamine-Cu NPs as a catalyst for the beta-borylation of alpha,beta-unsaturated ketones and ester, providing alkylboronate esters in up to 98% yield. Reuse of the magnetically recyclable catalyst resulted in no significant loss of activity in up to five reaction runs for both systems.

A chemoselective cascade cycloaddition reaction is developed for green and efficient access to cyclopenta-[c]pyridine derivatives. Simple and inexpensive NaOH is used as the sole catalyst for this process. The delta-carbon of cyclopropyl ester is activated as a nucleophiic carbon to initiate highly chemoselective cascade reactions. Cyclopenta-[c]pyridines bearing various substituents are afforded in excellent yields. Preliminary studies on the bioactivities of the afforded products show promising antibacterial activities for potential applications in plant protections.

Background: With respect to the increase in the average life expectancy, Alzheimer Disease (AD), the most common form of age-related dementia, has become a major threat to the population over the age of 65 during the past several decades. The majority of AD treatments are focused on cholinergic and amyloid hypotheses. Objective: In this study, three series of diphenyl-2-(2-(4-substitutedpiperazin-l-yl)ethyl)pyridazin-3(2H)-one derivatives were designed, synthesized and investigated for their ability to inhibit both cholinesterase enzymes and amyloid-beta aggregation. Method: The inhibitory activities of the synthesized compounds on AChE (from electric eel) and BChE (from equine serum) were determined by the modified Ellman’s method. The reported thioflavin T-based fluorometric assay was performed to investigate the effect of the selected compounds on the aggregation of A beta(1-42). The cytotoxic effect of the compounds (4g, 11g and 18g) was monitored in 3T3 cell lines to gain insight into therapeutic potential of the compounds by using MTT assay. The crystal structures of the AChE (1EVE) and BChE (1POI) enzymes were retrieved from the RCSB Protein Data Bank and Molecular Operating Environment (MOE) software was used for molecular docking of the ligands. Results: Among the tested compounds, 5,6-diphenyl derivative 18g was identified as the most potent and selective AChE inhibitor (IC50 = 1.75 mu M, Selectivity Index for AChE > 22.857). 4,6-Diphenyl derivative 11g showed the highest and the most selectivity for BChE (IC50= 4.97 mu M, SI for AChE < 0.124). Interestingly, 4,5-diphenyl derivative 4g presented dual cholinesterase inhibition (AChE IC50= 5.11 mu M; BChE IC50= 14.16 mu M, SI for AChE = 2.771). Conclusion: Based on biological activity results and low toxicity of the compounds, it can be said that diphenyl substituted pyridazinone core is a valuable scaffold. Especially, dual inhibitory potencies of 4,5-diphenylpyridazin-3(2H)-one core for the cholinesterase enzymes and A beta-aggregation makes this core a promising disease-modifying agent. A series of 1,3,5-triazine analogues, incorporating aminobenzene sulfonamide, aminoalcohol/ phenol, piperazine, chalcone, or stilbene structural motifs, were evaluated as potential antioxidants. The compounds were prepared by using step-by-step nucleophilic substitution of chlorine atoms in starting 2,4,6-trichloro-1,3,5-triazine. Reactions were catalyzed by Cu(I)-supported on a weakly acidic resin. The radical scavenging activity was determined in terms of %inhibition activity and EC50, using the ABTS method. Trolox and ascorbic acid (ASA) were used as standards. In the lowest concentration 1 X 10(-4) M, the %inhibition activity values at 0 min were comparable with both standards at least for 10 compounds. After 60 min, compounds 5, 6, 13, and 25 showed nearly twice %inhibition (73.44-87.09%) in comparison with the standards (Trolox = 41.49%; ASA = 31.07%). Values of EC50 at 60 min (17.16-27.78 mu M) were 5 times lower for compounds 5, 6, 13, and 25 than EC50 of both standards (trolox = 178.33 mu M; ASA = 147.47 mu M). Values of EC50 correlated with %inhibition activity. Based on these results, the presented 1,3,5-triazine analogues have a high potential in the treatment of illnesses caused or related to oxidative stress.

Astragalus mongholicus (MG) and Astragalus membranaceus (MJ), both generally known as Huangqi in China, are two perennial herbals widely used in variety diseases. However, there were still some differences in the chemical ingredients between MG and MJ. In this paper, metabolomics combined with the ultra-high performance liquid chromatography coupled with electrospray ionization/quadrupole time-of-flight mass spectrometry (UHPLC-ESI-Q-TOF-MS/MS) was employed to contrastively analyze the chemical constituents between MG and MJ. As a result, principal component analysis showed that MG and MJ were separated clearly. A total of 53 chemical markers were successfully identified for the discrimination of MG and MJ. Of them, the contents of 36 components including Astragaloside IIII, Astragaloside IV, Agroastragaloside I, etc. in MJ were significantly higher than those in MG. On the contrary, the contents of 17 other components including coumaric acid, formononetin, sophoricoside, etc. in MG were obviously higher than those in MJ. The results showed that the distinctive constituents in MG and MJ were remarkable, and MJ may own stronger pharmacological activities than MG. In a word, MG and MJ may be treated as two different herbs. This paper demonstrated that metabolomics was a vitally credible technology to rapidly screen the characteristic chemical composition of traditional Chinese medicine.

The catalytic asymmetric epoxidation of alpha,beta-unsaturated ketones by tert-butylhydroperoxide (TBHP) has been well established using rare-earth metal amides RE[N(SiMe3)(2)](3) (RE = La(1), Nd(2), Sm(3), Y(4), Yb(5)) with chiral TADDOL ligands. It was found that the combination of Yb[N(SiMe3)(2)](3) and chiral TADDOL ligand H2L2 ((4S,5S)-2,2-dimethyl-alpha,alpha,alpha ‘,alpha ‘-tetra-3,5-bis(trifluormethylphenyl)-1,3-dioxolane-4,5-dimethanol) in a 1 : 1 molar ratio was the optimal choice, which provided the desired epoxides in excellent yields (89-99%) and good to high enantioselectivities (57-94% ee), using DBU as an additive. Various substrates were proved to have functional group tolerance. In addition, gram-level experiments and derivatization experiments were also studied.
