A highly enantioselective three-component hydroacyloxylation/1,4-conjugate addition ofortho-hydroxybenzyl alcohols, ynamides and carboxylic acids was developed under mild reaction conditions in the presence of a chiralN,N ‘-dioxide/Sc(OTf)(3)complex, which went throughin situgeneratedortho-quinone methides with alpha-acyloxyenamides, delivering a range of corresponding chiral alpha-acyloxyenamides derivatives containinggem(1,1)-diaryl skeletons in moderate to good yields with excellenteevalues. The scale-up experiment and further derivation showed the practicality of this catalytic system. In addition, a possible catalytic cycle and transition state model was proposed to elucidate the origin of the stereoselectivity based on X-ray crystal structure of the alpha-acyloxyenamide intermediate and product.

Magnetite nanoparticles (MNPs) have been evaluated for inhibiting microbial growth and biofilm formation. In this study the effect of the nanocomposite Moroccan propolis extract / MNPs acting against methicillin resistant strains of Staphylococcus aureus (MRSA) was evaluated. Chemical composition of propolis was established by pyrolysis coupled to gas chromatography and mass spectrometry method (pyrolysis GC/MS). MNPs were obtained through the co-precipitation method. The fabricated nanostructure was characterized by X-ray Diffraction (DRX), Transmission Electron Microscopy (TEM), and Fourier Transform-Infrared Spectroscopy (FTIR). TEM of MNPs provided a particle average size of 15 nm, FTIR spectral analysis enabled a fast way of identification of MNPs, attesting the occurrence of the different combinations. The use of MNPs loaded with propolis and the antibiotic chloramphenicol at Minimum Inhibitory Concentration (MIC) value inhibited the bacterial growth of MSSA (methicillin susceptible strain of S. aureus) and MRSA strains. After the treatment with MNPs-OA-P-CLo nanocomposite (MNPs with oleic acid, propolis and chloramphenicol), the disruption of the bacterial cell was observed by TEM. The combination of propolis and chloramphenicol in free form at MIC value largely impaired both MSSA and MRSA strains as, after 2 h of treatment, no viable cells of MRSA 2 and MRSA 16 were recovered. Hence, the results elucidated a new antibacterial nanocomposite synthesis, for possible applications as prospective nanoantibacterial agents or drug carriers. A new and simple method for the deoxygenation of ketones to alkanes is presented. Most substrates are reduced under mild conditions by triethylsilane in the presence of catalytic amounts of [Rh(mu-Cl)(CO)(2)](2). This system selectively provides the methylene hydrocarbons in good to excellent yields starting from acetophenones and diaryl ketones. A rapid examination of the reaction pathway suggests that the ketone is first converted into an alcohol, which then undergoes hydrogenolysis to give the alkane.

Highly efficient alkylation of beta-chloro ketones and their derivatives was achieved by means of domino dehydrochlorination/Mn-enabled radical-based alkyl-alkyl cross-coupling reaction. Sulfonyl chlorides are inexpensive reactants extensively explored for functionalization, but never considered for radical hydrosulfonylation of alkenes. Herein, we report that tris(trimethylsilyl)silane is an ideal hydrogen atom donor enabling highly effective photoredox-catalyzed hydrosulfonylation of electron-deficient alkenes with sulfonyl chlorides. To increase the generality of this transformation, polarity-reversal catalysis (PRC) was successfully implemented for alkenes bearing alkyl substituents. This late-stage functionalization method tolerates a remarkably wide range of functional groups, is operationally simple, scalable, and allows access to building blocks which are important for medicinal chemistry and drug discovery.

A general and concise method was developed for the synthesis of 2-amino-4,6-diarylpyridine derivatives 4-6 through the cascade reaction, which includes Michael addition, intramolecular cyclization, aromatization, and/or loss of HNO2, of different types of alpha,beta-unsaturated ketones 1 and 1,1-enediamines 2 and 3 in 1,4-dioxane promoted by the base Cs2CO3 or piperidine. This method is suitable for the efficient parallel synthesis of pyridines. A library of highly functional 2-amino-4,6-diarylpyridine derivatives was easily constructed using the cascade reaction described in this study.

In an attempt to synthesise new tyrosinase inhibitors, we designed and synthesised a series of chalcone-hydroxypyridinone hybrids as potential tyrosinase inhibitors adopting strategic modifications of kojic acid. All the newly synthesised compounds were characterised by NMR and mass spectrometry. Initial screening of the target compounds demonstrated that compounds1a,1d, and1nhad relatively strong inhibitory activities against tyrosinase monophenolase, with IC(50)values of 3.07 +/- 0.85, 2.25 +/- 0.8 and 2.75 +/- 1.19 mu M, respectively. The inhibitory activity against monophenolase was 6- to 8-fold higher than that of kojic acid. Compounds1a,1d, and1nalso showed inhibition of diphenolase, with IC(50)values of 17.05 +/- 0.07, 11.70 +/- 0.03 and 19.3 +/- 0.28 mu M, respectively. The inhibition kinetics of diphenolase indicates that compounds1aand1dinduce reversible inhibition on tyrosinase. Finally, we found that copper coordination should be one of the important inhibitory mechanism of these compounds in tyrosinase.

The complex [Cp*RuCl(COD)] reacts with LH2Cl2 (L = bis(3-methylimidazol-2-ylidene)) and LiBun in tetrahydrofuran at 65 degrees C furnishing the bis-carbene derivative [Cp*RuCl(L)] (2). A thermomorphic ionic-liquid-based microemulsion system was successfully applied for the Ru-catalyzed asymmetric transfer hydrogenation of ketones. On the basis of the temperature-dependent multiphase behavior of the targeted microemulsion, simple product separation as well as catalyst recycling could be realized. The use of water-soluble ligands improved the immobilization of the catalyst in the microemulsion phase and significantly decreased the catalyst leaching into the organic layer upon extraction of the product. Eventually, the optimized microemulsion system could be applied to a wide range of aromatic ketones that were reduced with good isolated yields (up to 98%) and enantioselectivities (up to 97%), while aliphatic ketones were less successful.

The synthesis of different metal pincer complexes coordinating to the chiral PNP ligand bis(2-((2R,5R)-2,5-dimethyl-phospholanoethyl))amine is described in detail. The characterized complexes with Mn, Fe, Re and Ru as metal centers showed good activities regarding the reduction of several prochiral ketones. Comparing these catalysts, the non-noble metal complexes produced best selectivities not only for aromatic substrates, but also for different kinds of aliphatic ones leading to enantioselectivities up to 99% ee. Theoretical investigations elucidated the mechanism and rationalized the selectivity.
