An efficient approach to prepare trifluoromethyl-alpha-carbolines and ester group-substituted alpha-carbolines via the tandem cyclization reaction of 2-(2-aminophenyl)acetonitriles and trifluoromethyl 1,3-diones or beta,gamma-unsaturated alpha-ketoesters was reported. The transformation proceeded smoothly in the presence of catalytic environmental-benign iron salts, which are used to prepare the desired products in moderate to good yields.

Aim: There is a continuous and urgent need for new anticancer agents with novel structures and target selectivity. Methods & results: The anticancer activity of the prepared compounds was assessed against human lung (A549) and stomach (AGS) cancer cell lines and evaluated in the noncancer human lung fibroblast (MRC-5) cell line. 2-Pyrazolines were devoid of toxicity in all cell lines used, chalcones bearing a beta-(benz)imidazole moiety being toxic toward AGS cell line. Mechanistic studies showed that these compounds trigger loss of cell viability and mitochondrial membrane potential, while eliciting morphological traits compatible with regulated cell death, which was ultimately shown to derive from caspase activation, specifically caspase-3. Conclusion: Chalcones 1-3 have been identified as new and promising anticancer agents toward the AGS cell line.

We herein report highly chemoselective transfer hydrogenation of alpha,beta-unsaturated carbonyl compounds to saturated carbonyls with formic acid as a hydrogen donor over a stable and recyclable heterogeneous Pd nano particles (NPs) on N,O-dual doped hierarchical porous biomass-derived carbon. The synergistic effect between Pd NPs and incorporated heteroatoms on carbon plays a critical role on promoting the reaction efficiency. A series of alpha,beta-aromatic and aliphatic unsaturated carbonyl compounds was selectively reduced to their corresponding saturated carbonyls in up to 97% isolated yields with good tolerance of various functional groups. In addition, the catalyst can be successively reused for at least 6 times without significant loss in reaction efficiency.

In spite of the fact that a variety of reactions have been exploited for creation of innumerable porous organic polymers (POPs), aldol condensation reactions between aldehydes and ketones leading to enones remain unutilized quite inexplicably. We surmised that the enone functionality can be exploited for stabilization of Pd akin to the manner of Pd(0) in Pd-2(dba)(3) by developing POPs based on aldol condensation reactions of polycarbonyl compounds with dialdehydes, and that such materials can be employed for catalytic transformations. By subjecting rationally designed tri-/tetraacetyl-functionalized aryl amines to aldol condensations with terephthalaldehyde, three different POPs, i.e., TPAPOPs 1-3, that feature enone functionalities have been synthesized and shown to exhibit palpable gas sorption properties. Remarkably, inverse uptake for sorption of H-2 over N-2 was observed for all POPs. As surmised, the representative POP, i.e., TPAPOP-1, was found to stabilize in situ-generated Pd(0) nanoparticles to enable application of the resultant material, i.e., Pd@TPAPOP-1, as a recyclable heterogeneous catalyst for a number of organic transformations. It is shown that coupling reactions such as Suzuki and Heck, and reductions such as nitro-to-amine and hydrogenation of olefins can be accomplished in a facile manner by employing Pd@TPAPOP-1 as a heterogeneous recyclable catalyst; the aforementioned transformations have been demonstrated on a broad set of substrates for each type. Tyrosinase is an important component of the enzyme polyphenol oxidase, which upon contact with the phenolic substrates forms the pigment melanin and induces undesirable food browning. The phenolic and triterpenoid compounds that naturally occur in plants are well known as tyrosinase inhibitors. Combretum micranthum (CM) leaves, Euphorbia hirta (EH) plant, and Anacardium occidentale (AO) fruits are traditionally known to have potential anti-tyrosinase activities. The aim of this study was to optimize the ultrasound-assisted extraction of secondary metabolites from these matrices, and to evaluate in tubo the antityrosinase activity of these extracts. Efforts were also taken to profile the secondary metabolites, mainly the phenolic and triterpenoid compounds, in order to understand their probable association with tyrosinase inhibition. The optimal ultrasound-assisted extraction conditions for simultaneous extraction of phenolic, and triterpenoid compounds were determined. The aqueous fraction of these extracts showed significant antityrosinase activity, with the CM leaves exhibiting the strongest inhibitory effect (IC50 of 0.58 g.L-1). The predominant metabolic compounds from these natural extracts were putatively identified by using a high-resolution quadrupole-time of flight (QToF) LC-MS instrument. The high-resolution accurate mass-based screening resulted in identification of 88 predominant metabolites, which included dihydrodaidzein-7-O-glucuronide, micromeric acid, syringic acid, morin, quercetin-3-O-(6 ”-malonyl-glucoside), 4-hydroxycoumarin, dihydrocaffeic acid-3-O-glucuronide, to name some, with less than 5 ppm of mass error.

One-step synthesis of beta-carboxyl ketones from alpha,beta-unsaturated ketones using CO2 as a feedstock by photoelectro-synergetic catalysis is reported herein. In this ambient system, silica nanowires (SiNWs) electrode not only acts as the working electrode but also harvests extra light energy to promote the reaction. Bis(NHC) ligand precursors, L1, based on trans-1,2-diaminocyclohexane were designed and synthesized. To introduce chirality at the hydroxyamide side arm on the NHC of L1, a chiral beta-amino alcohol, such as enantiopure leucinol, was used. Cu-catalyzed asymmetric conjugate addition reactions of cyclic and acyclic enones with Et2Zn were selected to evaluate the performance of L1 as a chiral ligand. For the reaction of cyclic enone, a combination of [bis(trimethylsilyl)acetylene]-(hexafluoroacetylacetonato)copper(I) (Cu(hfacac)(btmsa)) with a (+/-)-trans-1,2-cyclohexanediamine-based bis(NHC) ligand precursor, (rac; S,S)-L1, which was prepared from (S)-leucinol, was the most effective. Thus, treating 2-cyclohexen-1-one (3) with Et2Zn in the presence of catalytic amounts of Cu(hfacac)(btmsa) and (rac; S,S)-L1 afforded (R)-3-ethylcyclohexanone ((R)-4) with 97% ee. Similarly, use of (rac; R,R)-L1, which was prepared from (R)-leucinol, produced (S)-4 with 97% ee. Conversely, for the asymmetric 1,4-addition reaction of the acyclic enone, optically pure (-)-trans-1,2-cyclohexanediamine-based bis(NHC) ligand precursor, (R,R; S,S)-L1, worked efficiently. For example, 3-nonen-2-one (5) was reacted with Et 2 Zn using the CuOAc/(R,R; S,S)-L1 catalytic system to afford (R)-4-ethylnonan-2-one ((R)-6) with 90% ee. Furthermore, initially changing the counterion of the Cu precatalyst between an OAc and a ClO4 ligand on the metal reversed the facial selectivity of the approach of the substrates. Thus, the conjugate addition reaction of 5 with Et2Zn using the Cu(ClO4)(2)/(R,R; S,S)-L1 catalytic system, afforded (S)-6 with 75% ee.

An inexpensive, mild, and highly efficient epoxidation protocol has been developed involving bubbling SO2F2 gas into a solution of olefin, 30% aqueous hydrogen peroxide, and 4 N aqueous potassium carbonate in 1,4-dioxane at room temperature for 1 h with the formation of the corresponding epoxides in good to excellent yields. The novel SO2F2/H2O2/K2CO3 epoxidizing system is suitable to a variety of olefinic substrates including electron-rich and electron-deficient ones.

Two chiral polymers based on binaphthol(BINOL) were synthesized. The polymer can be used as a chiral inducer to coordinate with ZnEt2 to form a recoverable self-supporting catalyst, which could be used in the asymmetric catalytic reaction of alpha, beta-unsaturated ketones. The target products of high yield and good enantioselectivity(e.e. value up to 99%) can be obtained under mild conditions. The catalytic activity was not reduced significantly after 4 or 5 cycles of recovery of the recovered polymers.
