UV-activated alkyne-alkene [2+2] cycloaddition has served as an important tool to access cyclobutenes. Although broadly adopted, the limitations with UV light as an energy source prompted us to explore an alternative method. Here we report alkyne-alkene [2+2] cycloaddition based on visible light photocatalysis allowing the synthesis of diverse cyclobutenes and 1,3-dienes via inter- and intramolecular reactions. Extensive mechanistic studies suggest that the localized spin densities at sp(2) carbons of alkenes account for the productive sensitization of alkenes despite their similar triplet levels of alkenes and alkynes. Moreover, the efficient formation of 1,3-dienes via tandem triplet activation of the resulting cyclobutenes is observed when intramolecular enyne cycloaddition is performed, which may serve as a complementary means to the Ru(II)-catalyzed enyne metathesis. In addition, the utility of the [2+2] cycloaddition has been demonstrated by several synthetic transformations including synthesis of various extended pi-systems.

A one-pot method for joining three separate components leading to an assortment of 4-arylquinazolines (27 examples) in good to excellent yields is described. The method consists of a palladium(II)-catalyzed- cascade reaction involving C(sp)-C(sp(2)) coupling followed by intramolecular C-N bond formation. Heteroarylation of alkenes with aryl iodides was efficiently achieved with a (MeDalphos)AuCl complex through Au-I/Au-III catalysis. The possibility to combine oxidative addition of aryl iodides and yr-activation of alkenes at gold is demonstrated for the first time. The reaction is robust and general (> 30 examples including internal alkenes, 5-, 6-, and 7-membered rings). It is regioselective and leads exclusively to trans addition products. The (P,N) gold complex is most efficient with electron-rich aryl substrates, which are troublesome with alternative photoredox/oxidative approaches. In addition, it provides a very unusual switch in regioselectivity from 5-exo to 6-endo cyclization between the Z and E isomers of internal alkenols.

We report the highly enantioselective synthesis of P-chiral tertiary phosphine oxides featuring an ethynyl group via Cu(i)-catalyzed azide-alkyne cycloaddition. Newly developed chiral pyridinebisoxazolines (PYBOX) bearing a bulky C4 shielding group play an important role in achieving excellent enantioselectivity while suppressing side bis-triazoles formation in desymmetrizing prochiral diethynylphosphine oxides. Notably, by tuning the size of the C4 shielding group, it is possible to achieve excellent remote enantiofacial control in desymmetrizing phosphole oxide-diynes with the prochiral P-center farther from the ethynyl group by four covalent bonds. Time-dependent enantioselectivity is observed for these desymmetric CuAAC reactions, suggesting a synergic combination of a desymmetrization and a kinetic resolution, and our ligands prove to be better than unmodified PYBOX in both steps. This finding contributes to a highly enantioselective kinetic resolution of racemic ethynylphosphine oxides. The resulting chiral ethynylphosphine oxides are versatile P-chiral synthons, which can undergo a number of diversifying reactions to enrich structural diversity.

The chalcone derivatives 3a-d reacted with either malononitrile or ethyl cyanoacetate in ethanol in the presence of catalytic amount of ammonium acetate in an oil bath at 120 degrees C to give the Knowevenagel condensation products 5a-h. The latter compounds reacted with hydrazine hydrate and afforded the 4H-pyrazole derivatives 7a-h, respectively. The reaction of compounds 7a-h with ethyl cyanoacetate in dimethylformamide under refluxing condition afforded the cyanoacetamido derivatives 8a-h, respectively. When compounds 8a-h reacted with elemental sulfur and either of malononitrile or ethyl cyanoacetate in ethanol containing triethylamine, the thiophene derivatives 9a-h and 10a-h, respectively, were obtained. The structure of the newly synthesized compounds was established by the analytical and spectral data. All the newly synthesized compounds were evaluated against the six cancer cell lines: A549, HT-29, MKN-45, U87MG, and SMMC-7721 and H460. Compounds 3c, 5h, 7g, 7h, 8f, 9e, 9g, and 10g were selected to examine their Pim-1 kinase inhibition activity as these compounds showed high inhibition toward the c-Met kinase and the tested cancer cell lines. Furthermore, compounds 3b, 3c, 5g, 5h, 7f, 7g, 7h, 8e, 8f, 8g, 8h, 9e, 9f, 9g, 9h, 10g and 10h were selected to be tested for pan-assay interference compounds analysis (PAINS). Almost all the tested compounds showed zero PAINS alert and can be used as drug compounds in the future.

With raising prevalence of obesity, the regulation of human body fat is increasingly relevant. The modulation of fatty acid uptake by enterocytes represents a promising target for body weight maintenance. Recent results demonstrated that the trigeminal active compounds capsaicin, nonivamide, and trans-pellitorine dose-dependently reduce fatty acid uptake in differentiated Caco-2 cells as a model for the intestinal barrier. However, non-pungent alternatives have not been investigated and structural determinants for the modulation of intestinal fatty acid uptake have not been identified so far. Thus, based on the previous results, we synthesized 23 homovanillic acid esters in addition to the naturally occurring capsiate and screened them for their potential to reduce intestinal fatty acid uptake using the fluorescent fatty acid analog Bodipy-C12 in differentiated Caco-2 cells as an enterocyte model. Whereas pre-incubation with 100 mu M capsiate did not change fatty acid uptake by Caco-2 enterocytes, a maximum inhibition of -47% was reached using 100 mu M 1-methylpentyl-2-(4-hydroxy-3-methoxy-phenyl)acetate. Structural analysis of the 24 structural analogues tested in the present study revealed that a branched fatty acid side chain, independent of the chain length, is one of the most important structural motifs associated with inhibition of fatty acid uptake in Caco-2 enterocytes. The results of the present study may serve as an important basis for designing potent dietary inhibitors of fatty acid uptake.

A method has been developed for the in situ sulfidation of Pd/C under continuous flow. The approach provides a cheap, sustainable, and operationally convenient method for chemoselective conjugate reduction by continuous hydrogenation. High conversions and excellent selectivities were obtained for olefin reduction in alpha/beta-unsaturated carbonyl compounds in the presence of hydrogenatively sensitive functionalities. The methodology was analyzed with a green metric system to highlight the sustainability features of the process.

Synthesis of fifteen C-4-aroyl-C-5-aryl-N-1-(5 ‘-deoxythymidin-5 ‘-yl)-1,2,3-triazoles have been reported starting from azidation of 5 ‘-p-toluenesulfonyloxythymidine followed by azide-alkene oxidative cycloaddition reaction of the resulted 5 ‘-azido-5 ‘-deoxythymidine with 1,3-diarylpropenones in dimethylformamide (DMF) in the presence of tetra-n-butylammonium hydrogen sulfate (n-Bu4N+HSO4-, TBAHS) as catalyst in 60 to 79% overall yields. Further, they were also synthesized by one pot sequential reaction of tosylated thymidine with sodium azide in DMF and then with 1,3-diarylpropenones in presence of n-Bu(4)N(+)HSO(4)(-)in superior yield of 70 to 95% than 60 to 79% in two step procedure. All fifteen synthesized compounds were screened for theirin vitroantiMycobacterium tuberculosisactivity against sensitive reference strain H37Rv and multi drug resistant (MDR) clinical isolate 591, and found to exhibit minimum inhibitory concentration (MIC) ranging from 2 to 15 mu g/mL, which was equivalent to the MIC of first line anti-tubercular drug streptomycin. All compounds qualify for their drug likeness when their physicochemical parameters were assessed using online MolSoft and Lipinski filter software, except their molecular weight. The cytotoxicity of potent compounds evaluated human monocytic cell line THP-1 by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay was found to be less as compared to the first line drug, isoniazid.
