A transition metal free expedient approach for the C=C bond cleavage of electron deficient alkenes such as arylidene Meldrum’s acid and malononitrile derivatives are discussed. The C=C bond of these compound were cleaved to benzoic acid in good yield at high temperature. Most importantly, with oxone in CH3CN/H2O at 45 degrees C or m-CPBA in DCM or NaClO2 in THE/H2O or PIDA in THE at room temperature furnished benzaldehyde derivatives selectively in excellent yields. The regioselective hydroboration of aliphatic internal alkenes remains a great challenge. Reported herein is an iridium-catalyzed hydroboration of aliphatic internal alkenes, providing distal-borylated products in good to excellent yields with high regioselectivity (up to 99:1). We also demonstrate that the C-B bond of the distal-borylated product can be readily converted into other functional groups. DFT calculations indicate that the reaction proceeds through an unexpected Ir-III/Ir-V cycle.

The condensation of 4H-1,2,4-triazol-3-amine with ortho esters derived from acetic, propionic, and benzoic acids gave the corresponding N-(4H-1,2,4-triazol-3-yl) carboximidates which were treated with cyanamide, carbon disulfide, and sodium thiocyanate to afford 7-substituted [1,2,4]triazolo[4,3-a][1,3,5]triazin-5-amines, [1,2,4]triazolo[4,3-c][1,3,5]thiadiazine-5-thiones, and [1,2,4]triazolo[4,3-c][1,3,5]thiadiazin-5-imines, respectively. The structures of the synthesized compounds were confirmed by IR, H-1 and C-13 NMR, and mass spectra and elemental analyses.

A panel of six new arene Ru (II)-NHC complexes 2a-f, (NHC = 1,3-diethyl-(5,6-dimethyl)benzimidazolin-2-ylidene 1a, 1,3-dicyclohexylmethyl-(5,6-dimethyl)benzimidazolin-2-ylidene 1b and 1,3-dibenzyl-(5,6-dimethyl)benzimidazolin-2-ylidene 1c) were synthesized from the transmetallation reaction of Ag-NHC with [(eta(6)-arene)RuCl2](2) and characterized. The ruthenium (II)-NHC complexes 2a-f were developed as effective catalysts for alpha-alkylation of ketones and synthesis of bioactive quinoline using primary/amino alcohols as coupling partners respectively. The reactions were performed with 0.5 mol% catalyst load in 8 h under aerobic condition and the maximum yield was up to 96%. Besides, the different alkyl wingtips on NHC and arene moieties were studied to differentiate the catalytic robustness of the complexes in the transformations.

A series of novel chalcone derivatives containing a thiophene sulfonate group were designed and synthesized. The structures of all title compounds were determined by H-1-NMR, C-13-NMR and HRMS. Antibacterial bioassays indicated that, compound 2l demonstrated excellent antibacterial activities against Xanthomonas axonopodis pv. citri (Xac), with an EC50 value of 11.4 mu g mL(-1), which is significantly superior to those of bismerthiazol (BT) (51.6 mu g mL(-1)) and thiodiazole-copper (TC) (94.7 mu g mL(-1)). Meanwhile, the mechanism of action of compound 2l was confirmed by using scanning electron microscopy (SEM). In addition, compound 2e showed remarkable inactivation activity against Tobacco mosaic virus (TMV), with an EC50 value of 44.3 mu g mL(-1), which was superior to that of ningnanmycin (120.6 mu g mL(-1)). Microscale thermophoresis (MST) also showed that the binding of compounds 2e and 2h to Tobacco mosaic virus coat protein (TMV-CP) yielded K-d values of 0.270 and 0.301 mu mol L-1, which are better than that of ningnanmycin (0.596 mu mol L-1). At the same time, molecular docking studies for 2e and 2h with TMV-CP (PDB code: ; 1EI7) showed that the compound was embedded well in the pocket between the two subunits of TMV-CP in each case. These results suggested that chalcone derivatives containing a thiophene sulfonate group may be considered as activators in the design of antibacterial and antiviral agents.

The development of an N-heterocyclic carbene (NHC) catalysed intercepted dehomologation of aldoses is reported. The unique selectivity of NHCs for aldehydes is exploited in the complex context of reducing sugars. Examples of strong substrate governance for either intercepted dehomologation or a subsequent redox-lactonisation were identified and mechanistically understood. More importantly, it was shown that catalyst design allowed the tuning of the selectivity of the reaction with structurally unbiased starting materials towards either of the two scenarios.

A highly selective and efficient oxidative carbocyclization/borylation of enallenols catalyzed by palladium immobilized on amino-functionalized siliceous mesocellular foam (Pd-AmP-MCF) was developed for diastereoselective cyclobutenol synthesis. The heterogeneous palladium catalyst can be recovered and recycled without any observed loss of activity or selectivity. The high diastereoselectivity of the reaction is proposed to originate from a directing effect of the enallenol hydroxyl group. In this work, we describe the development of the rearrangement for 7-aryl-substituted oxazolo[S,4-b]pyridines treated with aluminum chloride into synthetically hard-to-reach benzo[c][1,7]naphthyridinones. The discovered rearrangement is applied to a variety of electron-rich (hetero)arene substrates. It offers the advantages of mild conditions (90 degrees C temperature), fast reaction rates (<4 h), compatibility with air moisture, and the use of inexpensive commercial reagents. The proposed reaction mechanism and key elementary reaction acts were studied in detail using quantum chemical calculations. The feasibility of using activity-directed synthesis to drive antibacterial discovery was investigated.
