Oleanolic acid induces the Type III secretion system of ralstonia solanacearum

Ralstonia solanacearum, the causal agent of bacterial wilt, can naturally infect a wide range of host plants. The type III secretion system (T3SS) is a major virulence determinant in this bacterium. Studies have shown that plant-derived compounds are able to inhibit or induce the T3SS in some plant pathogenic bacteria, though no specific T3SS inhibitor or inducer has yet been identified in R. solanacearum. In this study, a total of 50 different compounds were screened and almost half of them (22 of 50) significantly inhibited or induced the T3SS expression of R. solanacearum. Based on the strong induction activity on T3SS, the T3SS inducer oleanolic acid (OA) was chosen for further study. We found that OA induced the expression of T3SS through the HrpG-HrpB pathway. Some type III effector genes were induced in T3SS inducing medium supplemented with OA. In addition, OA targeted only the T3SS and did not affect other virulence determinants. Finally, we observed that induction of T3SS by OA accelerated disease progress on tobacco. Overall our results suggest that plant-derived compounds are an abundant source of R. solanacearum T3SS regulators, which could prove useful as tools to interrogate the regulation of this key virulence pathway.

The Crystal Structures of Three Rhodanine-3-Carboxylic Acids

Abstract: The rhodanine derivatives show various pharmacological activities. Rhodanine-3-carboxylic acids can be used as the substrates in various synthesis of compounds containing rhodanine-3-carboxyalkyl moiety. In this paper new crystal structures of rhodanine-3-acetic acid and its two homologues, i.e. rhodanine-3-propionic acid and rhodanine-3-butyric acid, are reported. The relationship between the length of the alkyl chain and the geometry of these molecules was studied. The crystal network is dominated by strong hydrogen bonds O?H¡¤¡¤¡¤O formed by the carboxyl groups. Additionally, weak C?H¡¤¡¤¡¤O and C?H¡¤¡¤¡¤S contacts are observed. Graphical Abstract: To study the difference in intermolecular interactions of rhodanine-3-carboxylic acid, three crystal structures were determined by X-ray diffraction method. The crystal network in all studied structures is built of homosynthons and stabilized by weak C?H¡¤¡¤¡¤O and C?H¡¤¡¤¡¤S contacts.[Figure not available: see fulltext.]

Morpholine is a heterocycle featured in numerous approved and experimental drugs as well as bioactive molecules. It is often employed in the field of medicinal chemistry for its advantageous physicochemical, biological, and metabolic properties, as well as its facile synthetic routes. The morpholine ring is a versatile and readily accessible synthetic building block, it is easily introduced as an amine reagent or can be built according to a variety of available synthetic methodologies. This versatile scaffold, appropriately substituted, possesses a wide range of biological activities. There are many examples of molecular targets of morpholine bioactive in which the significant contribution of the morpholine moiety has been demonstrated; it is an integral component of the pharmacophore for certain enzyme active-site inhibitors whereas it bestows selective affinity for a wide range of receptors. A large body of in vivo studies has demonstrated morpholine’s potential to not only increase potency but also provide compounds with desirable drug-like properties and improved pharamacokinetics. In this review we describe the medicinal chemistry/pharmacological activity of morpholine derivatives on various therapeutically related molecular targets, attempting to highlight the importance of the morpholine ring in drug design and development as well as to justify its classification as a privileged structure.

Syntheses and Crystal Structures of Benzyl Substituted Thiazolidin-2-cyanamide Derivatives

Abstract: Reaction of thiazolidin-2-cyanamide and substituted benzyl bromide compounds in acetonitrile at room temperature afforded the 3-(2?-substituted benzyl)thiazolidin-2-cyanamide derivatives 1?13 in good yields. Compounds 1?13 were characterized by proton nuclear magnetic resonance (1H NMR) and infrared spectroscopies, of which the structures of the isomeric o-, m-, and p-fluoro derivatives 4?6 were established by single crystal X-ray crystallography. Compound 4 crystallizes in the monoclinic space group P21/n, with a = 9.177(19), b = 8.551(18), c = 14.090(3) A, beta = 98.243(3), and Z = 4. The unit cell of 5 has a monoclinic P21/c symmetry with the cell parameters a = 9.289(2), b = 14.057(4), c = 8.574(2) A, beta = 100.350(3), and Z = 4. The unit cell of 6 also has a monoclinic P21/c symmetry with the cell parameters a = 9.333(4), b = 14.034(5), c = 8.508(3) A, beta = 99.15(5), and Z = 4. Studies on aromatic schiff bases from methyl-1-naphthyl ketone. Part-I: Synthesis and characterization of ketimines from 1-acetylnaphthalene with derivatives of aniline

Schiff bases(Ketimines) were prepared from Methyl-1-naphthyl-ketone with 2-Hydroxy-aniline, 3-Hydroxyaniline, 4-Hydroxy-aniline, 3-Nitroanilines and 4-Bromo-aniline using toluene as solvent by azeotropic(reflux) method using Dean and Stark. The synthesized ketimines were characterized by colour, physical constants, TLC and FTIR spectra. The purity of the synthesized compounds was confirmed from the information gathered from TLC, elemental and FTIR spectral analysis.
