Fluxional chemical species such as bullvalene have been a valuable source of inspiration and fundamental insight into the nature of chemical bonds. A supramolecular analogue of bullvalene, i.e., a fluxional host-guest system, in which the ensemble of a well-defined host and guest is engaged in continuous, degenerate constitutional rearrangements, is still elusive, however. Here, we report experimental and computational evidence for guest-induced dynamic covalent rearrangements in the ammonium complexes of self-assembled orthoester cryptands. This unique behavior is made possible by the ammonium guest playing a dual role: it is sufficiently acidic to initiate dynamic covalent exchange reactions at the orthoester bridgeheads, and as a hydrogen bond donor it acts as a supramolecular template, governing the outcome of a multitude of possible intra- and intermolecular rearrangement reactions. One particularly striking example of inherent dynamic behavior was observed in host-guest complex [Na-4(+)subset of o-Me-2-2.1.1], which spontaneously rearranged into the larger and thermodynamically more stable complex [NH4+subset of o-Me-2-2.2.1], even though this process led to the formation of poor host o-Me-2-1.1.1 as a consequence of the excess of one subcomponent (diethylene glycol; 1 in our nomenclature). These inherently adaptive host-guest networks represent a unique platform for exploring the interrelationship between kinetic and thermodynamic stability. For instance, as a result of optimal NH4+ binding, complex [NH4+subset of o-Me-2-2.2.1] was found to be thermodynamically stable (negligible intermolecular rearrangements over weeks), whereas computational studies indicate that the compound is far from kinetically stable (intramolecular rearrangements).

One-step synthesis of beta-carboxyl ketones from alpha,beta-unsaturated ketones using CO2 as a feedstock by photoelectro-synergetic catalysis is reported herein. In this ambient system, silica nanowires (SiNWs) electrode not only acts as the working electrode but also harvests extra light energy to promote the reaction. Following a possible two-step single-electron-transfer process, various substrates exhibit good efficiency with up to 98% product yield, and the method shows wide functional group tolerance.

Reduced graphene oxide (rGO)-NaBH4 is reported as mild and efficient catalyst-system for chemo-/ regioselective reduction of structurally different aliphatic, aromatic as well as alpha,beta-unsaturated aldehydes and ketones in water. The rGO was prepared by reducing graphene oxide using Tulsi leaf extract as bio-reductant. Operational simplicity, ambient reaction condition, high yield of pure products (80-97%), no by-product formation, no use of column chromatography for purification are the salient features of the envisaged protocol. A one-pot route to 2-alkyl and 2-aryl-4H-benzo[d][1,3]oxazin-4-ones (also known as 4H-3,1-benzoxazin-4-ones) has been developed and studied. The method involves the reaction of aryl-substituted anthranilic acids with orthoesters in ethanol catalyzed by acetic acid. Additionally, we have also investigated the reaction under microwave conditions. Not all of the substrates were successful in yielding the target heterocycles as some of the reactions failed to undergo the final elimination. This process led to the isolation of (+/-)-2-alkyl/aryl-2-ethoxy-1,2-dihydro-4H-benzo[d][1,3]oxazin-4-ones. The formation of the dihydro analogs correlated with the electron density on the aromatic ring: Electron-donating groups favored the 4H- benzo[d][1,3]oxazin-4-ones, while electron-withdrawing groups tended to favor the dihydro product. Substituting a pyridine ring for the benzene ring in the substrate acid suppressed the reaction.

This strategy incorporates a radical acyl thiol-ene reaction as the key C-S bond forming step. Cyclization is achieved via a Steglich-type thiolactonization of 5-mercaptopentanoic acids. We report the facile and scalable synthesis of delta-thiolactones in moderate to good yield under mild reaction conditions with tolerance for a range of functional groups.

In search for new molecular entities as anti-TB agents, the benzimidazoquinazoline polyheterocyclic scaffold has been designed adopting the scaffold hopping strategy. Thirty-two compounds have been synthesized through an improved tandem decarboxylative nucleophilic addition cyclocondensation reaction of o-phenylenediamine with isatoic anhydride followed by further cyclocondensation of the intermediately formed 2-(o-aminoaryl) benzimidazole with trialkyl orthoformate/acetate. The resultant benzimidazoquinazolines were evaluated in vitro for anti-TB activity against M. tuberculosis H37Rv (ATCC27294 strain). Fourteen compounds exhibiting MIC values in the range of 0.4-6.25 mu g/mL were subjected to cell viability test against RAW 264.7 cell lines and were found to be non-toxic (< 30% inhibition at 50 mu g/mL). The active compounds were further evaluated against INH resistant Mtb strains. The most active compound 6x [MIC (H37Rv) of 0.4 mu g/mL] and the compound 6d [MIC (H37Rv) of 0.78 mu g/mL] were also found to be active against INH resistant Mtb strain with MIC values of 12.5 and 0.78 mu g/mL, respectively. In this article, a direct synthetic method for alkyl 5-arylthiophene-2-thiocarboxylates was reported. Treatment of alkyl 2-aroyl-1-chlorocyclopropanecarboxylates with excess amount of Lawesson’s reagent readily afforded alkyl 5-arylthiophene-2-thiocarboxylates in good to excellent yields under mild conditions. This cycloisomerization reaction process would be initiated by a ring-opening of the strained cyclopropane.

Novel Mannich base derivatives of glabridin were synthesized and their antiproliferative activity were performed along with our previously reported glabridin-chalcone hybrids molecules (GCHMs) against various human cell lines MDA-MB-231 (breast adenocarcinoma), HEK-293 (embryonic kidney cell line), K562 (leukemia), MCF-7 (breast adenocarcinoma), HeLa (cervix adenocarcinoma), HepG2 (hepatocellular carcinoma) and WRL-68 (hepatic carcinoma). The result showed that the glabridin significantly reduced cell proliferation with IC(50)ranges from 3.67 to 58.30 mu M against all the tested cell lines. The remarkable reduction in antiproliferative activity 2′,4′-dimethoxyglabridin and GCHMs compounds with phenolic OH groups protected by methoxy (OCH3) groups suggested that the free OH groups are essential factor for the antiproliferative activity of glabridin and its derivatives. The Mannich base derivatives of glabridin showed moderate activity IC50(2.20->95.78 mu M). Furthermore,in silicotarget identification analysis revealed that AKT1, DECR1 and NOS1 are the potential targets for glabridin and their derivatives.

Bacterial lipopolysaccharides (LPS) are important bio-medical structures, playing a major role in the interaction with human immune systems. Their core regions, containing multiple units ofl-glycero-d-mannoheptoses (l,d-heptose), are highly conserved structurally (withO3 andO7 glycosidic bonds), making them an epitope of high interest for the potential development of new antibiotics and vaccines. Research in this field has always been restricted by the limited availability of the parentl,d-heptose as well as its biochemical epimeric precursord-glycero-d-mannoheptose (d,d-heptose). This problem of availability has recently been solved by us, through a rapid and efficient practical synthesis ofl,d-manno-heptose peracetate demonstrated at scale. Herein we report an optimized, technically simple and versatile synthetic strategy for the differentiation of both thel-glyceroandd-glycero-d-mannoheptose scaffolds. Our approach is based on an orthoester methodology for the differentiation of all three positions of the sugar core using aO6,O7-tetraisopropyl disiloxyl (TIPDS) protecting group for the exocyclic positions. Furthermore, the regioselective opening toward 7-OH acceptors (6O-FTIPDS ethers) differentiates the exocyclic diol which has been demonstrated with a broader set of substrates and for bothmanno-heptoses for the first time.
