An efficient and sustainable protocol for the hydration of alkynes has been developed under metal/acid/catalyst/ ligand-free conditions in a water/ethyl lactate mixture. The hydrogen-bond network in the ethyl lactate and water mixture plays a crucial and decisive role in activating the alkynes for hydration to afford the corresponding methyl ketones. This strategy gives the Markovnikov (ketone) addition product selectively over other possible products. The essential role of hydrogen bonding has been confirmed by experimental and theoretical techniques. A probable mechanism has been suggested by various control tests. The efficacy of the method has been further explored for the competent production of value-added alpha,beta-unsaturated carbonyl compounds through the reaction of aldehydes with alkynes as ketonic surrogates. The environmentally benign hydration method takes place under mild conditions, has broad functional-group compatibility, and uses the ethyl lactate/water (1:3) medium as a green alternative in the absence of any hazardous, harmful, or expensive substances. A new methodology for the synthesis of alpha-diketones was reported via a one-pot cascade process from aldehydes and ketones catalyzed by a bifunctional iron nanocomposite using H2O2 as a green oxidant in water. The catalytic asymmetric epoxidation of alpha,beta-unsaturated ketones by tert-butylhydroperoxide (TBHP) has been well established using rare-earth metal amides RE[N(SiMe3)(2)](3) (RE = La(1), Nd(2), Sm(3), Y(4), Yb(5)) with chiral TADDOL ligands. It was found that the combination of Yb[N(SiMe3)(2)](3) and chiral TADDOL ligand H2L2 ((4S,5S)-2,2-dimethyl-alpha,alpha,alpha ‘,alpha ‘-tetra-3,5-bis(trifluormethylphenyl)-1,3-dioxolane-4,5-dimethanol) in a 1 : 1 molar ratio was the optimal choice, which provided the desired epoxides in excellent yields (89-99%) and good to high enantioselectivities (57-94% ee), using DBU as an additive. Various substrates were proved to have functional group tolerance. In addition, gram-level experiments and derivatization experiments were also studied.

Two series of [1,2,4]triazolo[4,3-a]pyrazine derivatives bearing 4-oxo-pyridazinone moieties (compounds 21a-l and 22a-l) were designed and their IC50 values were evaluated against three cancer cell lines (A549, MCF-7 and HeLa) and c-Met kinase. Among them, the compound with most potential, 22i, exhibited excellent anti-tumor activity against A549, MCF-7 and HeLa cancer cell lines with IC50 values of 0.83 +/- 0.07 mu M, 0.15 +/- 0.08 mu M and 2.85 +/- 0.74 mu M, respectively, and it also possessed superior c-Met kinase inhibition ability at the nanomolar level (IC50 = 48 nM). Moreover, dose-dependent experiments, AO fluorescence staining, cell cycle assay, Annexin V-FITC/PI staining and docking studies were carried out in this study. The results demonstrated that compound 22i could be a potential c-Met kinase inhibitor.

Olefin aminations are important synthetic technologies for the construction of aliphatic C-N bonds. Here we report a catalytic protocol for olefin hydroamidation that proceeds through transient amidyl radical intermediates that are formed via proton-coupled electron transfer (PCET) activation of the strong N-H bonds in N-alkyl amides by an excited-state iridium photocatalyst and a dialkyl phosphate base. This method exhibits a broad substrate scope, high functional group tolerance, and amenability to use in cascade polycyclization reactions. The feasibility of this PCET protocol in enabling the intermolecular anti-Markovnikov hydroamidation reactions of unactivated olefins is also demonstrated.

Several hundred (396) compounds from New Zealand flora with medicinal properties were analyzed for their physicochemical properties. It was found that approximately 10% fulfilled all the requirements to be considered to be lead-like, over half of the compounds were deemed to be in the drug-like space and approximate to 75% were in the known drug space. These results indicate the presence of a significant proportion of compounds that are of particular interest to pursue as potential lead compounds or therapeutics. Additionally, compound classes were analyzed separatelymost carbonyl-containing compounds (aldehydes, ketones, esters and lactones), along with phenols were the most lead-like compounds, which also displayed very good proportions in the drug-like and known drug space. The information presented herein can be harnessed and utilized in future work, through focussing on the compounds and compound classes that exhibit high-levels of lead-likeness for further development.

A new and simple method for the deoxygenation of ketones to alkanes is presented. Most substrates are reduced under mild conditions by triethylsilane in the presence of catalytic amounts of [Rh(mu-Cl)(CO)(2)](2). This system selectively provides the methylene hydrocarbons in good to excellent yields starting from acetophenones and diaryl ketones. A rapid examination of the reaction pathway suggests that the ketone is first converted into an alcohol, which then undergoes hydrogenolysis to give the alkane.
