An asymmetric synthesis of two analogues of SL0101 (1) has been achieved. The effort is aimed at the discovery of inhibitors of the p90 ribosomal S6 kinase (RSK) with improved bioavailability. The route relies upon the use of the Taylor catalyst to regioselectively install C-3 ” acetyl or carbamate functionality. This study led to the identification of a third-generation analogue of SL0101 with a C-4 ” n-Pr-carbamate and a C-3 ” acetate with improved RSK inhibitory activity.

Chitosan microspheres modified by 2-pyridinecarboxaldehyde were prepared and used in the construction of a heterogeneous catalyst loaded with nano-Cu prepared by a reduction reaction. The chemical structure of the catalyst was investigated by Fourier Transform Infrared Spectroscopy (FT-IR), Scanning Electron Microscopy (SEM), Transmission Electron Microscopy (TEM), and X-ray Photoelectron Spectroscopy (XPS). Under mild conditions, such as no ligand at room temperature, the catalyst was successfully applied to catalyze the borylation of alpha,beta-unsaturated receptors in a water-methanol medium, yielding 17%-100% of the corresponding beta-hydroxy product. Even after repeated use five times, the catalyst still exhibited excellent catalytic activity.

The effect of Argentine propolis extracts against enzymes related to metabolic syndrome and oxidative stress, as well as the botanical origin of raw propolis, were studied. Histological and chemical analyses of propolis samples revealed that the botanical origin isZuccagnia punctata, an Argentine medicinal plant. The melissopalynological analysis showed both pollen grains ofZ. punctataand the other plant species. This result indicates that the differences found in the botanical remains compared to the palynological studies may have been caused by the bees selecting resinous shrubs mainly ofZ. punctatafor the production of propolis and other plants with flowers for the production of honey. The richness of propolis was remarkable in two flavonoid precursors (2 ‘,4 ‘-dihydroxy-3 ‘-methoxychalcone, 2 ‘,4 ‘-dihydroxychalcone), the major chemical components ofZ. punctata. The hydroalcoholic extracts of Argentine propolis andZ. punctatawere able to inhibit enzymes associated with the metabolic syndrome, including alpha-glucosidase, alpha-amylase and lipase, with IC(50)values between 7 to 14, 37 to 48, and 13 to 28 mu g/mL, respectively. Biological activity was mainly attributed to chalcones. Oxygen and nitrogen reactive species scavenging activity was determined by the assays of superoxide radical (O-2(center dot-)), hydroxyl radical (HO center dot), hydrogen peroxide (H2O2), nitric oxide (NO center dot), and cation radical (ABTS(center dot+)). Results showed SC(50)values between 115 to 278, 12.50 to 46; 39 to 92; 50 to 104.50 and 23 to 33.75 mu g/mL, respectively. This study suggests for the first time that propolis from Argentina is highly effective in inhibiting enzymes related to the metabolic syndrome and in free-radical scavenging that would justify its use as a dietary supplement or as a functional ingredient in special food. Practical Application Propolis from Catamarca, Argentina, is traditionally used as medicine and food. Its botanical origin isZuccagnia punctata, an endemic plant species popularly used as a medicine in Argentina. Propolis has the ability to regulate the activity of enzymes involved in the carbohydrate and lipid metabolism, and consequently in metabolic syndrome. Besides, its antioxidant capacity makes it a natural product that can be used as a dietary supplement or as a functional ingredient in special foods. It is important to highlight that in the Argentine Food Code, propolis was incorporated in 2008 as a dietary supplement and the present results give major added value to this product.

Single-atom photocatalysis as an important organic transformation strategy has received increasing attention, with the performances largely depending on the design of catalysts. This protocol involves initially the fabrication of a single-atom photocatalyst Ni/TiO2 for the visible-light-induced site-selective sulfonation of enamides to give amidosulfones with 36 examples up to 99% yield. The experimental results show that this single-atom photocatalyst Ni/TiO2 can achieve site-selective sulfonation of enamide to construct alpha-amidosulfones and beta-propionamidosulfones under visible light. Importantly, such a single-atom photocatalysis-based synthesis system exhibits favorable recyclability, high turnover number (up to 18 963), excellent tolerance of functional groups, and can be easily scaled up with good efficiency.

Traditional metal implants such as titanium, cobalt, and chromium have found wide utility in medicine; however, these come with a risk of toxicity. To overcome metal-related toxicity and enable degradability, polyesters including polycaprolactone (PCL), polylactic acid (PLA), and polyglycolic acid (PGA) show promise for the replacement of various biomedical applications of metals due to their accepted biocompatibility and FDA approval. However, polyesters are less stiff than their metallic counterparts, limiting their application to non-load bearing injury sites, such as fixation hardware for fingers. To improve mechanical properties, graphene oxide (GO)-polyester composites are a promising class of biodegradable scaffolds. Initial reports of these composites are encouraging, but mechanical properties still fall short. Traditional composites rely on non-covalent association between GO and the polyesters, which often leads to failure at the interface and weakens the overall strength of the material. Herein, we present a strategy for attachment of these FDA-approved polyesters onto a derivative of GO using a robust covalent bond. By covalently functionalizing the graphenic backbone with polyesters and without metal catalysts, we create functional graphenic materials (FGMs) to not only simultaneously retain biodegradability and compatibility, but also mechanically strengthen PCL, PLA, and PGA; we observed an average increase in the Young’s modulus of over 140% compared to the graphenic backbone. These polyester-functionalized FGMs are a promising platform technology for tissue implants.

UV-activated alkyne-alkene [2+2] cycloaddition has served as an important tool to access cyclobutenes. Although broadly adopted, the limitations with UV light as an energy source prompted us to explore an alternative method. Here we report alkyne-alkene [2+2] cycloaddition based on visible light photocatalysis allowing the synthesis of diverse cyclobutenes and 1,3-dienes via inter- and intramolecular reactions. Extensive mechanistic studies suggest that the localized spin densities at sp(2) carbons of alkenes account for the productive sensitization of alkenes despite their similar triplet levels of alkenes and alkynes. Moreover, the efficient formation of 1,3-dienes via tandem triplet activation of the resulting cyclobutenes is observed when intramolecular enyne cycloaddition is performed, which may serve as a complementary means to the Ru(II)-catalyzed enyne metathesis. In addition, the utility of the [2+2] cycloaddition has been demonstrated by several synthetic transformations including synthesis of various extended pi-systems.

Direct selective desaturation of carbonyl compounds to synthesize alpha,beta-unsaturated carbonyl compounds represents an environmentally benign alternative to classical stepwise procedures. In this study, we designed an ideal CeO2-supported Pd(II)-on-Au nanoparticle catalyst (Pd/Au/CeO2) and successfully achieved heterogeneously catalyzed selective desaturation of cyclohexanones to cyclohexenones using O-2 in air as the oxidant. Besides cyclohexenones, various bioactive enones can also be synthesized from the corresponding saturated ketones under open air conditions in the presence of Pd/Au/CeO2. Preliminary mechanistic studies revealed that alpha-C-H bond cleavage in the substrates is the turnoverlimiting step of this desaturation reaction.

Agave lechuguilla is one of the most abundant species in arid and semiarid regions of Mexico, and is used to extract fiber. However, 85 % of the harvested plant material is discarded. Previous bioprospecting studies of the waste biomass suggest the presence of bioactive compounds, although the extraction process limited metabolite characterization. This work achieved flavonoid profiling of A. lechuguilla in both processed and non-processed leaf tissues using transcriptomic analysis. Functional annotation of the first de novo transcriptome of A. lechuguilla (255.7 Mbp) allowed identifying genes coding for 33 enzymes and 8 transcription factors involved in flavonoid biosynthesis. The flavonoid metabolic pathway was mostly elucidated by HPLC-MS/MS screening of alcoholic extracts. Key genes of flavonoid synthesis were higher expressed in processed leaf tissues than in non-processed leaves, suggesting a high content of flavonoids and glycoside derivatives in the waste biomass.
