Thiazolidinone and its derivatives have high pharmacological relevance since they are available in both natural products and Pharmaceutical compounds. The main synthetic routes to thiazolidinones comprising three components such as an amine, a carbonyl group and mercapto acid. The classical method of synthesis reported may be either a one-pot three-component condensation method or a two-step process. Synthesis and anticancer activity evaluation of thiazolidinones containing benzothiazole moiety. These compounds were screened for in-vitro anticancer activity. The activity data exhibits that all compounds were found to show potent anticancer activity. Various substituents at C-2 and C-3 of thiazolidinone results in potent anticancer activity. Prompted by these reports, we aimed to prepare the following series of 2, 3-disubstituted-Thiazolidinone derivatives as potent anticancer agents.

Preparation, antibacterial evaluation and preliminary structure-activity relationship (SAR) study of benzothiazol- and benzoxazol-2-amine derivatives

In this study, a novel benzothiazol- and benzooxazol-2-amine scaffold with antibacterial activity was designed and synthesized. Preliminary structure-activity relationship analysis displayed that compound 8t with a 5,6-difluorosubstituted benzothiazole was found to be a potent inhibitor of Gram-positive pathogens, and exhibited some potential against drug-resistant bacteria and without cytotoxicity in therapeutic concentrations. In addition, molecular docking studies indicated that Staphylococcus aureus methionyl-tRNA synthetase might be the possible target of these compounds. Taken together, the present study provides an effective entry to the synthesis of a good lead for subsequent optimization and a new small molecule candidate drug for antibacterial therapeutics.

Antioxidants and chronic obstructive pulmonary disease

Antioxidants represent an attractive therapeutic avenue for individuals with chronic obstructive pulmonary disease (COPD). Cigarette smoke, the major cause of COPD, contains very high concentrations of gaseous and soluble oxidants that can directly induce cell injury and death. Furthermore, particulate matter in cigarette smoke activates lung macrophages that subsequently attract neutrophils. Both neutrophils and macrophages from the lungs of cigarette smokers continuously release large amounts of superoxide and hydrogen peroxide through the nicotinamide adenine dinucleotide phosphate (NADPH) oxidase complex. Once individuals with COPD stop smoking, the neutrophilic inflammation in the airways and lung parenchyma persists, as do the markers of oxidative stress. Several animal models of cigarette smoke-induced injury have provided evidence that various antioxidants may prevent inflammation and morphological changes associated with COPD however, evidence of benefit in patients is less abundant. Although oxidants can inactivate alpha-1 antitrypsin and other protective proteins, damage lung tissue, and increase mucus production, they also are essential for killing pathogens and resolving inflammation. This review will examine the pre-clinical and clinical evidence of a role for antioxidants in the therapy of patients with COPD.

Recent advances in the discovery of hif- 1alpha-p300/cbp inhibitors as anticancer agents

Hypoxia-inducible factor-1 (HIF-1), a heterodimeric (containing alpha and beta subunits) transcription factor, is involved in hypoxia response pathway that regulates the expression of many tumor-related genes. The stabilized HIF-1 heterodimer couples to the general co-activators p300/CBP (CREB binding protein), forming an active transcription factor to initiate hypoxic responses. Inhibiting the transcription factor-coactivator HIF-1alpha-p300/CBP interaction represents an attractive approach for blocking hypoxia pathway in tumors. Recently, diverse HIF-1alpha-p300/CBP inhibitors have been designed and their anti-tumor activities have been evaluated. The developments of inhibitors of HIF-1alpha- p300/CBP are discussed in this review. An outline of structures and biological activities of these inhibitors can be traced, along with the approaches for inhibitors discovery. The challenges in identifying novel and selective potent inhibitors of HIF-1alpha-p300/CBP are also put forward.
