Further studies of regioselective alkoxydehalogenation of 2,4-dichloroquinolines, 2,6-dichloropyridine and 2,4-dichloronitrobenzene

Regioselective alkoxydehalogenation reactions using solid alkoxide in toluene have been studied. 2-Alkoxy-4-haloquinolines were obtained from 2,4-dichloroquinolines. With 2,6-dichloropyridine, alpha-regioselectivity occurred to furnish the 2-alkoxy-6-chloropyridine. The reaction failed with 2,4-dichloronitrobenzene indicating that alkoxide surface contact with a basic heterocyclic nitrogen lone pair was essential for success. Comparative studies with the standard alkoxydehalogenation reaction (alcoholic alkoxide solution) have been performed, all compounds have been identified by 1H and 13C NMR.

Antiinflammatory activity of triazine thiazolidinone derivatives: Molecular docking and pharmacophore modelling

Some 3-(4,6-dichloro-1,3,5-triazin-2-yl)-2-phenylthiazolidin-4-one derivatives were prepared by cyclo-condensation reaction between 2-amino-4,6-dichloro-1,3,5-triazine, substituted aromatic aldehyde and ethyl-2-mercaptoacetate, with an yield in the range 76-86 %. Prepared compounds showed antiinflammatory activity. The halogenated electron-withdrawing groups on the phenyl ring of 4-thiazolinone generated antiinflammatory activity. Among the synthesized compounds, 3-(4,6-dichloro-1,3,5-triazin-2-yl)-2-(2,5-difluorophenyl)thiazolidin-4-one showed better antiinflammatory activity with 72 and 79 % inhibition for TNF-alpha and IL-6, respectively. Also, molecular docking and pharmacophore modelling performed for this active antiinflammatory compound highlighted that hydrophobicity as an important feature for activity optimization.

Asymmetric transfer hydrogenation of ketones using Ru(0) nanoparticles modified by Chiral Thiones

The catalytic asymmetric transfer hydrogenation (ATH) of acetophenone in isopropanol by Ru(0) nanoparticles (NPs) obtained by the in-situ reduction of Ru (II) half-sandwich complexes of chiral 2-oxazolidinethiones and 2-thiozolidinethiones was examined and compared with the catalytic activity of Ru(0) NPs formed in-situ by the reduction of [Ru(p-cymene)(Cl)2]2 in presence of optically active ligands such as (S)-4-isobutylthiazolidine-2-thione, (S)-4-Isopropyl-2(?2-pyridinyl)-2-oxazoline, (8S, 9R)-(?)-cinchonidine, (S)-leucinol, (S)-phenylalaninol, and (S)-leucine. Three of the best catalytic systems were then examined for ATH of thirteen aromatic ketones with different electronic and steric properties. A maximum of 24% ee was obtained using NPs generated from the Ru (II) half-sandwich complex with (S)-4-isobutylthiazolidine-2-thione in the TH of acetophenone. The NPs were characterized by TEM and DLS measurements. Kinetic studies and poisoning experiments confirmed that the reaction is catalyzed by the chiral NPs formed in-situ. Complete characterization of the complexes, including the X-ray crystallographic characterization of two complexes, was also carried out.

Synthesis of some 5-(substituted benzylidene)-3-phenyl-2-phenylimino-1,3-thiazolidin-4-ones under ultrasound irradiation

Recently, we have synthesized environmental benign one pot synthesis of 5-(Substituted Benzylidene)-3-Phenyl-2-Phenylimino-1,3-Thiazolidin-4-Ones (a-f) under Ultrasound irradiation by cyclization of 1,3 -diphenylthiourea with a-halo acids in presence of sodium acetate in absolute ethanol for 10-15 minute. The crude compound was purified by silica gel column chromatography using n-hexane and ethyl acetate as solvent system and recrystalised from ethanol. The synthesized compounds have been screened in vitro for their antimicrobial activity against S. aureus and E.coli as compare with standard drug Streptomycin. Some of the compounds displayed pronounced biological activity. The resulting products were characterized by IR,1H NMR and13 C NMR spectroscopic method.

Synthesis and biological evaluation of 4-thiazolidinone derivatives as antitubercular and antimicrobial agents

New series of N-[2-{2-(substitutedphenyl)-4-oxo-5-(substitutedbenzylidene)-1,3-thiazolidine}-iminoethyl]-2-amino-5-nitrothiazole, 5(a-m) have been synthesized from 2-amino-5-nitrothiazole as a starting material by conventional as well as microwave methods. All the synthesized compounds 4(a-m) were screened for their antibacterial and antifungal activities against some selected bacteria and fungi and antitubercular activity screened against Mycobacterium tuberculosis. The structure of all the synthesized compounds were confirmed by chemical and spectral analyses such as IR, 1H NMR, 13C NMR and FAB-Mass.

Six focused rhodanine-based libraries, 60 compounds in total, were synthesized and evaluated as potential dynamin I GTPase inhibitors. Twenty-six were more potent than the lead compound with 13 returning IC50 values ?10 muM, making the Rhodadyn series among the most active dynamin inhibitors reported. Two analogues were highly effective at blocking receptor-mediated endocytosis: C10 and D10 with IC50(RME) = 7.0 ¡À 2.2 and 5.9 ¡À 1.0 muM, respectively.
