Quantum chemical studies for some thiazolidinone derivatives using density functional theory

B3LYP/6-311G++(d, p) calculations have been carried out to study the structural and chemical properties of thioazilinone derivatives. The optimized molecular geometry of these derivative molecules are compared with the synthesized results of Mushtaque et al. The calculated results show that the density functional theory (DFT) can well reproduce the structure of the title compounds. Highest occupied-Lowest unoccupied molecular orbital (HOMO?LUMO) gap of the derivatives molecules are comparatively smaller then bare thiazolidinone. Natural bond orbital analysis for these title molecules shows strong delocalization of lone pair electrons from N, S, and O to the chemical bonds associated with central thiazolidinonering. The infrared intensites are calculated to compare with the FT-IR observation reported by Mushtaque et al. The experimental UV-Vis spectrum of present derivatives have been recorded and compared with calculated time dependent density functional (TDDFT) approach. The qualitative measures of chemical activeness of molecules have been presented in terms molecular electrostatic potential (MEP).

Dietary antioxidants prevent alcohol-induced ciliary dysfunction

Previously we have shown that chronic alcohol intake causes alcohol-induced ciliary dysfunction (AICD), leading to non-responsive airway cilia. AICD likely occurs through the downregulation of nitric oxide (NO) and cyclic nucleotide-dependent kinases, protein kinase G (PKG) and protein kinase A (PKA). Studies by others have shown that dietary supplementation with the antioxidants N-acetylcysteine (NAC) and procysteine prevent other alcohol-induced lung complications. This led us to hypothesize that dietary supplementation with NAC or procysteine prevents AICD. To test this hypothesis, C57BL/6 mice drank an alcohol/water solution (20% w/v) ad libitum for 6 weeks and were concurrently fed dietary supplements of either NAC or procysteine. Ciliary beat frequency (CBF) was measured in mice tracheas, and PKG/PKA responsiveness to beta-agonists and NOx levels were measured from bronchoalveolar lavage (BAL) fluid. Long-term alcohol drinking reduced CBF, PKG and PKA responsiveness to beta-agonists, and lung NOx levels in BAL fluid. In contrast, alcohol-drinking mice fed NAC or procysteine sustained ciliary function and PKG and PKA responsiveness to beta-agonists. However, BAL NO levels remained low despite antioxidant supplementation. We also determined that removal of alcohol from the drinking water for as little as 1 week restored ciliary function, but not PKG and PKA responsiveness to beta-agonists. We conclude that dietary supplementation with NAC or procysteine protects against AICD. In addition, alcohol removal for 1 week restores cilia function independent of PKG and PKA activity. Our findings provide a rationale for the use of antioxidants to prevent damage to airway mucociliary functions in chronic alcohol-drinking individuals.

Inhibiting the transcription factor-coactivator HIF-1alpha-p300/CBP interaction represents an attractive approach for blocking hypoxia pathway in tumors. Recently, diverse HIF-1alpha-p300/CBP inhibitors have been designed and their anti-tumor activities have been evaluated. The developments of inhibitors of HIF-1alpha- p300/CBP are discussed in this review. An outline of structures and biological activities of these inhibitors can be traced, along with the approaches for inhibitors discovery. The challenges in identifying novel and selective potent inhibitors of HIF-1alpha-p300/CBP are also put forward.

Thiazolidine prodrugs as protective agents against gamma-radiation-induced toxicity and mutagenesis in V79 cells

Representatives of two classes of thiazolidine prodrug forms of the well-known radioprotective agents L-cysteine, cysteamine, and 2-[(aminopropyl)amino]ethanethiol (WR-1065) were synthesized by condensing the parent thiolamine with an appropriate carbonyl donor. Inherent toxicity of the prodrugs was assessed in V79 cells using a clonogenic survival assay. Protection against radiation-induced cell death was measured similarly after exposure to 0-8 Gy gamma (137Cs) radiation. Antimutagenic activity was determined at the hypoxanthine-guanine phosphoribosyltransferase (HGPRT) locus. All thiazolidine prodrugs exhibited less toxicity than their parent thiolamines, sometimes dramatically so. Protection against radiation-induced cell death was observed for the 2-alkylthiazolidine, 2(R,S)-D-ribo-(1?,2?,3?,4?-tetrahydroxybutyl) thiazolidine (RibCyst), which produced a protection factor at 8 Gy of 1.8; the cysteine analogue, 2(R,S)-D-ribo-(1?,2?,3?,4?-tetrahydroxybutyl) thiazolidine-4(R)-carboxylic acid (RibCys), was less active. RibCyst also exhibited excellent antimutational activity, rivaling that of WR-1065. The 2-oxothiazolidine analogues showed little activity in either determination under the conditions tested, perhaps due to their enhanced chemical and biochemical stability.

Synthesis and in vitro anti-tumor activity of some new sebacoyl chloride based heterocycles

Background: N-Heterocycles are of special interest because, many natural and synthetic bioactive compounds are found to be N-containing heterocycles and they constitute an important class of pharmacophores in medicinal chemistry. Recently, 1,2,3-triazoles have been found to possess a vast range of vital applications in the agrochemical, pharmaceutical, and materials field. In addition, various compounds of the 1,2,3-triazole family have shown a broad spectrum of biological properties such as antibacterial, and anti-HIV activity. 1,3,4- Oxadiazoles are important oxygen and nitrogen containing heterocyclic compounds, they possess desirable electronic and charge-transport properties. Methods: Two mammalian cell lines were grown in RPMI-1640 medium, supplemented with 10% heat inactivated FBS, 50 units/mL of penicillin and 50 g/mL of streptomycin and maintained at 37 C in a humidified atmosphere containing 5% CO2. The cells were maintained as ?monolayer culture? by serial sub-culturing. Preliminary cytotoxicity was performed using SRB method. Results: Sebacoyl chloride and decanedihydrazide dihydrochloride are utilized as versatile building blocks to annulate a series of novel azole and/ or azine systems. The in vitro anti-tumor activities of the synthesized compounds were evaluated against human breast cancer cell line (MCF-7) and human cervical cancer cell line (HeLa). The obtained data indicated that the majority of the tested compounds possess significant anti-tumor activities against the tested tumor cell lines. Conclusion: Sebacoyl chloride, decanedihydrazide dihydrochloride and N-nucleophiles are useful precursors for the synthesis of different functionalized N- heterocycles. The cytotoxic study revealed that the majority of the tested compounds possess significant anti-tumor activity towards the tested tumor cell lines.
