Novel Mannich base derivatives of glabridin were synthesized and their antiproliferative activity were performed along with our previously reported glabridin-chalcone hybrids molecules (GCHMs) against various human cell lines MDA-MB-231 (breast adenocarcinoma), HEK-293 (embryonic kidney cell line), K562 (leukemia), MCF-7 (breast adenocarcinoma), HeLa (cervix adenocarcinoma), HepG2 (hepatocellular carcinoma) and WRL-68 (hepatic carcinoma). The result showed that the glabridin significantly reduced cell proliferation with IC(50)ranges from 3.67 to 58.30 mu M against all the tested cell lines. The remarkable reduction in antiproliferative activity 2′,4′-dimethoxyglabridin and GCHMs compounds with phenolic OH groups protected by methoxy (OCH3) groups suggested that the free OH groups are essential factor for the antiproliferative activity of glabridin and its derivatives. The Mannich base derivatives of glabridin showed moderate activity IC50(2.20->95.78 mu M). Furthermore,in silicotarget identification analysis revealed that AKT1, DECR1 and NOS1 are the potential targets for glabridin and their derivatives.

A diboron-mediated rhodium-catalysed transfer hydrogenation system using water as the hydrogen donor is developed. In addition to a series of alkenes with good functional group tolerance, this rhodium-based catalytic system also effectively reduces aldehydes and ketones. Direct selective desaturation of carbonyl compounds to synthesize alpha,beta-unsaturated carbonyl compounds represents an environmentally benign alternative to classical stepwise procedures. In this study, we designed an ideal CeO2-supported Pd(II)-on-Au nanoparticle catalyst (Pd/Au/CeO2) and successfully achieved heterogeneously catalyzed selective desaturation of cyclohexanones to cyclohexenones using O-2 in air as the oxidant. Besides cyclohexenones, various bioactive enones can also be synthesized from the corresponding saturated ketones under open air conditions in the presence of Pd/Au/CeO2. Preliminary mechanistic studies revealed that alpha-C-H bond cleavage in the substrates is the turnoverlimiting step of this desaturation reaction.

A highly efficient rongalite-mediated three-component radical annulation reaction to furnish fully substituted pyrazoles from aryldiazonium salts and alpha,beta-unsaturated aldehydes or ketones under metal- and oxidant-free conditions at room temperature has been developed. In this transformation, aryldiazonium salts served as the precursor of both the aryl and aryl hydrazine units. Mechanistic investigations indicated that rongalite could act as a radical initiator and reducing reagent simultaneously in the reaction.

Synthesis of fifteen C-4-aroyl-C-5-aryl-N-1-(5 ‘-deoxythymidin-5 ‘-yl)-1,2,3-triazoles have been reported starting from azidation of 5 ‘-p-toluenesulfonyloxythymidine followed by azide-alkene oxidative cycloaddition reaction of the resulted 5 ‘-azido-5 ‘-deoxythymidine with 1,3-diarylpropenones in dimethylformamide (DMF) in the presence of tetra-n-butylammonium hydrogen sulfate (n-Bu4N+HSO4-, TBAHS) as catalyst in 60 to 79% overall yields. Further, they were also synthesized by one pot sequential reaction of tosylated thymidine with sodium azide in DMF and then with 1,3-diarylpropenones in presence of n-Bu(4)N(+)HSO(4)(-)in superior yield of 70 to 95% than 60 to 79% in two step procedure. All fifteen synthesized compounds were screened for theirin vitroantiMycobacterium tuberculosisactivity against sensitive reference strain H37Rv and multi drug resistant (MDR) clinical isolate 591, and found to exhibit minimum inhibitory concentration (MIC) ranging from 2 to 15 mu g/mL, which was equivalent to the MIC of first line anti-tubercular drug streptomycin. All compounds qualify for their drug likeness when their physicochemical parameters were assessed using online MolSoft and Lipinski filter software, except their molecular weight. The cytotoxicity of potent compounds evaluated human monocytic cell line THP-1 by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay was found to be less as compared to the first line drug, isoniazid.

A unique 1,3-dipolar [3+2] cycloaddition of alkyl 4-oxo-4-arylbut-2-enoates bearing two different electron-withdrawing groups was completed by using the silver/(R)-DTBM-Segphos catalyst system, which gives the corresponding fully substituted pyrrolidines with four stereogenic centers in good yields and with excellent enantioselectivities (up to 98% ee).

Magnetite nanoparticles (MNPs) have been evaluated for inhibiting microbial growth and biofilm formation. In this study the effect of the nanocomposite Moroccan propolis extract / MNPs acting against methicillin resistant strains of Staphylococcus aureus (MRSA) was evaluated. Chemical composition of propolis was established by pyrolysis coupled to gas chromatography and mass spectrometry method (pyrolysis GC/MS). MNPs were obtained through the co-precipitation method. The fabricated nanostructure was characterized by X-ray Diffraction (DRX), Transmission Electron Microscopy (TEM), and Fourier Transform-Infrared Spectroscopy (FTIR). TEM of MNPs provided a particle average size of 15 nm, FTIR spectral analysis enabled a fast way of identification of MNPs, attesting the occurrence of the different combinations. The use of MNPs loaded with propolis and the antibiotic chloramphenicol at Minimum Inhibitory Concentration (MIC) value inhibited the bacterial growth of MSSA (methicillin susceptible strain of S. aureus) and MRSA strains. After the treatment with MNPs-OA-P-CLo nanocomposite (MNPs with oleic acid, propolis and chloramphenicol), the disruption of the bacterial cell was observed by TEM. The combination of propolis and chloramphenicol in free form at MIC value largely impaired both MSSA and MRSA strains as, after 2 h of treatment, no viable cells of MRSA 2 and MRSA 16 were recovered. Hence, the results elucidated a new antibacterial nanocomposite synthesis, for possible applications as prospective nanoantibacterial agents or drug carriers.
