Novel 2-(aryloxymethyl)aziridines and 2((3-aryl-1-phenylallyloxy)methyl)aziridine derivatives were prepared via ring-opening reaction of epoxides. The synthesized derivatives were characterized by using elemental analysis (EA), FT-IR, C-13 NMR, and H-1 NMR. The in vitro antitubercular activities of the synthesized compounds were evaluated against Mycobacterium tuberculosis H37Rv (MTB H37Rv) strain using MTT-MABA assay. All the aziridine derivatives exhibited improved persuasive antitubercular activity against MTB H37Rv in comparison with standard drugs. Among the tested compounds, 2-(naphthalene-1-yloxy) methyl aziridine (5b), 2-(naphthalene-2-yloxy)methylaziridine (5c), 2(m-tolyloxymethyl)aziridine (5e), 2-(3-(4-methoxyphenyl)-1-phenylalloxy)methylaziridine (12b) and 2-(3(2-chlorophenyl)-1-phenylallyloxy)methylaziridine (12c) revealed promising activity against MTh H37Rv. Specifically, compound 5b and 12 b showed three-times more active (MIC = 0.5 mu g/mL) than the standard drugs ethambutol (MIC = 1.56 mu g/mL) and ciprofloxacin (MIC = 1.56 mu g/mL). A highly enantioselective three-component hydroacyloxylation/1,4-conjugate addition ofortho-hydroxybenzyl alcohols, ynamides and carboxylic acids was developed under mild reaction conditions in the presence of a chiralN,N ‘-dioxide/Sc(OTf)(3)complex, which went throughin situgeneratedortho-quinone methides with alpha-acyloxyenamides, delivering a range of corresponding chiral alpha-acyloxyenamides derivatives containinggem(1,1)-diaryl skeletons in moderate to good yields with excellenteevalues. Indoles are one of the most valuable nucleophiles in Michael additions catalyzed by a proper Lewis acid. In this paper, we found that a cationic iron salt is effective to carry out the Michael addition of indoles. beta-Mono- and disubstituted enones reacted smoothly with indoles under our conditions. The cationic iron catalyst is very active, and the maximum TON was up to 425. Moreover, cationic iron-catalyzed conditions enabled a chemoselective Michael addition of a substrate possessing both enone and alpha,beta-unsaturated ester moieties.

Synthesis of fifteen C-4-aroyl-C-5-aryl-N-1-(5 ‘-deoxythymidin-5 ‘-yl)-1,2,3-triazoles have been reported starting from azidation of 5 ‘-p-toluenesulfonyloxythymidine followed by azide-alkene oxidative cycloaddition reaction of the resulted 5 ‘-azido-5 ‘-deoxythymidine with 1,3-diarylpropenones in dimethylformamide (DMF) in the presence of tetra-n-butylammonium hydrogen sulfate (n-Bu4N+HSO4-, TBAHS) as catalyst in 60 to 79% overall yields. Further, they were also synthesized by one pot sequential reaction of tosylated thymidine with sodium azide in DMF and then with 1,3-diarylpropenones in presence of n-Bu(4)N(+)HSO(4)(-)in superior yield of 70 to 95% than 60 to 79% in two step procedure. All fifteen synthesized compounds were screened for theirin vitroantiMycobacterium tuberculosisactivity against sensitive reference strain H37Rv and multi drug resistant (MDR) clinical isolate 591, and found to exhibit minimum inhibitory concentration (MIC) ranging from 2 to 15 mu g/mL, which was equivalent to the MIC of first line anti-tubercular drug streptomycin. All compounds qualify for their drug likeness when their physicochemical parameters were assessed using online MolSoft and Lipinski filter software, except their molecular weight. The cytotoxicity of potent compounds evaluated human monocytic cell line THP-1 by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay was found to be less as compared to the first line drug, isoniazid.

Orange peel oil contains high amount of d-limonene which is reported to have antidiabetic activity. Propolis contains many biologically active compounds like phenolic acids, flavonoids and terpenes. Because of its rich phenolic composition, antidiabetic activity of propolis has also been shown in literature. Consumption of raw propolis is limited because of its resinous nature. It should be extracted to convert it into consumable form. Ethanol is the most used solvent for this purpose. Ethanol is the limiting factor for propolis consumption either for religious concerns or being harmful for human health. There is an increasing need for new, compatible and healthier solvents for propolis extraction. This is the first paper describing the extraction of propolis with orange peel oil. 1:10 (g/v) ratio was used for extraction. Orange peel oil (OPO), orange peel oil propolis extract (OPOPE) and ethanol propolis extract (EPE) were separately analysed by using GC-MS technique. Total phenolic content of oil and propolis extract was also carried out. Inhibition of alpha-amylase from barley malt with new extract was also reported. It was found that OPOPE was rich in volatile compounds of propolis, phenolic acid derivatives and free fatty acids. It was observed that OPOPE showed higher inhibition on alpha-amylase enzyme (IC50 0.196 mg/mL). It can be concluded that orange peel oil could be used as solvent for preparing propolis extract. Propolis extract obtained with orange peel oil could also be used as complementary medicine for the treatment of type 2 diabetes.

Magnetite nanoparticles (MNPs) have been evaluated for inhibiting microbial growth and biofilm formation. In this study the effect of the nanocomposite Moroccan propolis extract / MNPs acting against methicillin resistant strains of Staphylococcus aureus (MRSA) was evaluated. Chemical composition of propolis was established by pyrolysis coupled to gas chromatography and mass spectrometry method (pyrolysis GC/MS). MNPs were obtained through the co-precipitation method. The fabricated nanostructure was characterized by X-ray Diffraction (DRX), Transmission Electron Microscopy (TEM), and Fourier Transform-Infrared Spectroscopy (FTIR). TEM of MNPs provided a particle average size of 15 nm, FTIR spectral analysis enabled a fast way of identification of MNPs, attesting the occurrence of the different combinations. The use of MNPs loaded with propolis and the antibiotic chloramphenicol at Minimum Inhibitory Concentration (MIC) value inhibited the bacterial growth of MSSA (methicillin susceptible strain of S. aureus) and MRSA strains. After the treatment with MNPs-OA-P-CLo nanocomposite (MNPs with oleic acid, propolis and chloramphenicol), the disruption of the bacterial cell was observed by TEM. The combination of propolis and chloramphenicol in free form at MIC value largely impaired both MSSA and MRSA strains as, after 2 h of treatment, no viable cells of MRSA 2 and MRSA 16 were recovered. Hence, the results elucidated a new antibacterial nanocomposite synthesis, for possible applications as prospective nanoantibacterial agents or drug carriers. The present study was planned to apply the nanotechnology to therapeutically proven flavonoid-rich fraction for obtaining a formulation with enhanced antihyperglycemic activity. The leaves were extracted in aqueous methanol and fractionated to yield a flavonoid-rich fraction. This fraction was formulated into an aqueous nanosuspension using PVA through the nanoprecipitation method. The developed formulations were characterized for physical parameters and optimum nanosuspension was evaluated against type 2 diabetes in high-fat diet-fed, streptozotocin-induced diabetic animals. The pharmacokinetic parameter and histopathology were assessed to determine the kinetic profile and tissue architectures post oral administration of selected nano suspension. The selected nanosuspension was also studied for stability under accelerated conditions. Flavonoidrich fraction was obtained using Kupchan partitioning. The developed nanosuspension PG-EA-NS-B showed optimum physical parameters. In hyperglycemia induced by streptozotocin, in high-fat diet-fed rats, it effectively restores the normal level of blood glucose in the first hour. The effects were comparable to standard Glibenclamide. In biochemical observations, PG-EA-NS-B, as well as PG-EA, showed beneficial effects on various hepatic and renal parameters. The pharmacokinetic parameters showed that nanosuspension enhances the absorption and decreases the metabolism of flavonoid actives. The selected nanosuspension and free flavonoid fraction showed rectification of histological changes. The nanosuspension was most stable in cool conditions. PVA based nanosuspensions can be a viable strategy for oral delivery of natural flavonoids, as it significantly affected the absorption and metabolism of flavonoids upon oral administration.

A unique 1,3-dipolar [3+2] cycloaddition of alkyl 4-oxo-4-arylbut-2-enoates bearing two different electron-withdrawing groups was completed by using the silver/(R)-DTBM-Segphos catalyst system, which gives the corresponding fully substituted pyrrolidines with four stereogenic centers in good yields and with excellent enantioselectivities (up to 98% ee).
